TheSMMC-7721cells of the test group were incubated in the RPMI-1640mediumwith different concentrations of remifentanil(0.001ã€0.01ã€0.1ã€1ã€10ã€100ã€200μmol/L)(R1-R7). Six parallel wells were set up in each groupand isopykniccell culture fluid was as blank control. Ultrastructure of the cells was observedby transmission electron microscopy. CCK-8method was used to detect cellproliferation activity when cells were incubated for24,48and72h respectively.Flow cytometry instrument was used to detect cell apoptosis and cell cycle. Theability of migration was assessed by scratches experiment.Results1. The cell of remifentanil group showed various degrees ofcellular swelling, chromatin broken and margination whereasthe shape of cellwas normal in control group.Membrane, nucleus and organelles of control cellwere complete.2. CCK-8method showed that72h average proliferation rate ofremifentanil group(R3-R7) cell were (0.917±0.032)%ã€(0.898±0.040)%ã€(0.861±0.057)%ã€(0.849±0.022)%ã€(0.853±0.036)%,which was significantly lower than the proliferation rate of the control group(1.121±0.051)%(P<0.05)when the concentration of remifentanil was over0.1μmol/L. However, nosignificant difference in cell proliferation was shown between100and200μmoL/L groups.3. When the concentration of remifentanil was over0.1μmol/L,remifentanil group(R3-R7)cell apoptosis rates were(20.8±0.98)ã€(27.6±1.21)%ã€(34.2±0.83)%ã€(38.8±0.97)%ã€(39.1±0.87)%,which were higher than the controlgroup(12.3±1.47)%(P<0.05). G0/G1phase increased and S phase decreasedwith the concentration increased(P<0.05).However, no significant difference incell cycle was shown between100and200μmoL/L groups.4. Remifentanilgroup(R1-R7)cell healing rate were (40.03±1.84)%ã€(34.51±1.79)%ã€(31.67±1.97)%ã€(25.87±1.49)%ã€(19.36±2.08)%ã€(12.40±2.14)%ã€(13.86±2.34)%,which were lower than the control group(44.37±2.57)%(P<0.05). However, nosignificant difference in cell cycle was shown between100and200μmoL/Lgroups.Conclusion When remifentanil was over0.1μmol/L, it can decrease humanhepatoma cell SMMC-7721proliferation, inhibit cell migration.
