| Objective:To observe the inhibitory effect on growth of hepatoma cells H22and the influence on immune function of Stichopus japonicus acid mucopolysaccharide(SJAMP) and fluorouracil (5-FU) combined with in Hepatocarcinoma22-bearing mouse by two part of experimental, exploring the anti tumor mechanisms of SJAMP; To investigate the attenuated and synergized effect of SJAMP in combination with5-FU on Hepatocarcinoma22-bearing mouse.Methods:Experiment1:The mice were injected H22cells into subcutaneous of the right armpit, then were randomly divided into blank control group (Normal Saline),5-FU group(20mg/kg BW), SJAMP low,mid,high dose groups (6.25,12.5,25mg/kg BW). Administered immediately after grouping.The mice were administered once a day via intraperitoneally inject continuously for12days. The inhibition rate of tumor, spleen indexes and thymus indexes were calculated. TNF-a, IL-2and IFN-β were detected by ELISA method. Neutral red method was used to detect macrophage phagocytosis. Spleen Lymphocytes proliferation capability was assayed by CCK-8method. NK cell function was detected by MTT method. T lymphocyte subsets were measured by flow cytometry.Experiment2:The mice were injected H22cells into subcutaneous of the right armpit, then were were randomly divided into5-FU group(20mg/kg BW), SJAMP group (25mg/kg BW), SJAMP+5-FU low-dose group(SLAMP25mg/kg BW and5-FU10mg/kg BW), SJAMP+5-FU high-dose group (SLAMP25mg/kg BW and5-FU20mg/kg BW). The other method ibid.Results:Experiment1:At the end of the experiment; The body weight in SJAMP groups increased significantly higher than that in5-FU group (P<0.05).The tumor weight in SJAMP groups were lower than those of blank control group, the SJAMP high dose group had significant difference (P<0.05).Compared with blank control group, the spleen index in SJAMP groups were higher, there were significant difference in the SJAMP mid dose group and SJAMP high dose group (P<0.05). The thymus index in SJAMP group were lower than that in blank control group, and there was no significant difference. The macrophage phagocytosis, spleen lymphocyte proliferation and NK cell function in SJAMP groups were higher than that of blank control group, there were statistical significance in SJAMP high dose group(P<0.05), and statistical significance on spleen lymphocyte proliferation and NK cell activity in SJAMP mid dose group (P<0.05). ELISA results showed that, compared with blank control group, serum TNF-a in SJAMP groups decreased significantly (P<0.05), the serum IL-2increased significantly in SJAMP mid dose group and SJAMP high dose group(P<0.05),the serum IFN-β increased significantly in SJAMP high dose group(P<0.05). The ratio of CD3+CD4+/CD3+CD8+in SJAMP groups were higher than that in blank control group (P<0.05).Experiment2:Compared with5-FU group, tumor weight was significantly decreased in SJAMP+5-FU high-dose group, The immune function of mice in combination groups had been obviously improved:in SJAMP+5-FU low-dose group, the spleen index, thymus index, abdominal cavity macrophage phagocytic function, spleen lymphocyte proliferation, NK cell activity, the level of serum IL-2, serum IFN-and the ratio of CD3+CD4+/CD3+CD8+were significantly increased (P<0.05); in SJAMP+5-FU high-dose group, macrophage phagocytic function, the proliferation of splenic lymphocyte, serum levels of IL-2and the ratio of CD3+CD4+/CD3+CD8+cells the ratio were significantly increased (P<0.05)Conclusion:1. SJAMP can inhibit the tumor growth in mice with H22liver cancer. And improve the immune function by promoting or maintaining the immune organs growth, enhancing the function of immune cells and regulating cytokine secretion and regulation T cells.2.Combined application of SJAMP and5-FU,Can improve the antitumor effect and reduce the side effect of5-FU, and improve the immune function of H22-bearing mouse. |
