| Kisspeptin is a kind of peptide encoded by KISS-1gene which mainly distributed in hypothalamus.It can promote the onset of puberty and play an important role in regulating the reproductive endocrinology by regulating the secretion of GnRH.Objective:To investigate the relationship between Kisspeptin and ovulation as well as ovrian function by detecting the concentration of Kisspeptin in serum of PCOSã€POFã€POI patients and serum or urine from different period of women menstrual cycle.Methods:1.The serum of PCOSã€POFã€POI patients and the serum and urine from different period of menstrual cycle in normal female without PCOSã€POF and POI(MC3dã€MC6dã€MC9dã€MClldã€MC12dã€MC13dã€MC14dã€MC16d) were collected.2.The concentration of Kisspeptin in different specimens were detected by ELISA.3.The variation tendency of the concentration of Kisspeptin in serum and urine as well as the relationship between Kisspeptin and sex hormone were analyzed by SPSS.Results:1.The concentration of Kisspeptin in serum of PCOSã€POFã€POI patients was higher than the control group(normal women).2.The variation of the concentration of Kisspeptin in a whole menstrual cycle was periodical.During the menstrual period,the concentration was very low and then it reached the first peak on the11th day (the size of follicle is about1.2cm), which was significantly higher than the menstrual phase(t=-8.769,P<0.001).During the13th day it reached the second peak which was lower than the first one.Then it declined until ovulation.3.The variation tendency of Kisspeptin in urine was the same with serum.It reached the first peak on the11th day and second peak appeared in the13th day.We also found a positive correlation between the concentration of Kisspeptin in urine and serum (R=0.879,P=0.001), which demonstrated that the variation of Kisspeptin in urine was the same with serum and can be used alternatively.4.The correlation analysis showed that there was no significant correlation in the concentration of Kisspeptin in serum with sex homone(LH and E2).But the correlation coefficient between Kisspeptin and E2which is three days after the Kisspeptin sample is high(R=0.797,P=P=0.413>0.01)Conclusion:1.Kisspeptin is related to many reproductive endocrine disease such as PCOSã€POIã€POF.The up-regulation of Kisspeptin has correlation with higher LH.This conclusion are consistent with Arreguin-Arevalo JA’s view.In other words,Kisspeptin is closely related to ovary function of female.Kisspeptin may predict the development of follicle and ovulation.2.The variation tendency of Kisspeptin in urine is the same with serum and there is a positive correlation.Therefore, the Kisspeptin of urine could replace serum in ovulation prediction.3. Kisspeptin may promote the secretion of E2. |
