The Expression Of RAC1and CDC42in Nasopharyngeal Carcinoma And Its Clinical Signification | | Posted on:2015-03-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q L Wu | Full Text:PDF | | GTID:2254330431453137 | Subject:Oncology | | Abstract/Summary: | PDF Full Text Request | | Objectice: In this study, immunohistochemistry was used to exploreRac1and Cdc42in the invasion and metastasis of NPC.Methods: Immunohistochemistry was utilized to detect the proteinexpression ratio and intensity of Rac1and Cdc42in172specimens of NPC ofdifferent clinical stages and20cases of nasopharyngeal mucosa chronicinflammation tissues. Transmission electron microscopy (TEM) was utilized toobserve the ultrastructural morphological changes of NPC cells which werepositive expression of Rac1, Cdc42.Results:1. The expression of Rac1protein and Cdc42protein in humanNPC was81.40%,83.72%. Both of them were significantly higher than thecontrol tissues (both0%). Statistically significant differences were exhibited(P<0.001).2. Positive expression of Rac1and Cdc42were related to TNMstages and clinical stages, there exhibited a statistically significant differenceamong each stages (P<0.01or P<0.05).3. The expression of Rac1and Cdc42 in NPC correlated significantly.(Rs=0.639, P=0.000).4. Median survival timeof patients with Rac1or Cdc42positive expression singly or their combinedexpression was significantly shorter than that with Rac1or Cdc42negativeexpression after treating.5. Transmission electron microscope reveals that thecarcinoma cells with positive expression of Rac1and Cdc42had obviouslymorphologic changes. Cytoplasmic membrane extended outside to generatethe pseudopodia, which are more and longer than these in control tissues. Conclusions: Nasopharyngeal carcinoma cells with positive expressionof Rac1and Cdc42had higher metastatic potential. The abnormal expressionof Rac1and Cdc42protein suggest that they probably play important roles inthe invasion and metastasis of NPC. Rac1and Cdc42may have a synergiceffect in invasion and metastasis of NPC. Rac1and Cdc42expression singly ortheir combined expression were significantly suggested poor prognosis. | | Keywords/Search Tags: | nasopharyngeal neoplasms, Rac1, Cdc42, immunohistochemistry, prognosis | PDF Full Text Request | Related items |
