| Objective: To explore the expression and function of intrathymicimmune-related miRNA and its target gene in myasthenia gravis patients withthymoma thymus.Methods: We collected13thymus near thymoma tissue of MGT group and13thymic tissue of normal control group(NC)and do microRNAs chip analysisto compare the miRNA profile of thymus tissue between the two groups, usinga combination of bioinformatics methods and microRNAs chip analysis result tofind immune-related microRNA and its target gene, using Q-PCR method anddual luciferase reporter system to analysis the regulatory relationships ofmicroRNA and its target gene.Results:1.The miRNA microarray chip analysis identified137miRNAsdifferentially expressed in thymoma tissue of myasthenia gravis compared withthe health control group, immune-related miR-125a-5p is one of mostup-regulated miRNAs(2.099579, P<0.01).2. By Bioinformatical analysis,FOXP3is target gene of miR-125a-5p.3.Q-PCR show that the expression ofFOXP3mRNA is downregulated and miR-125a-5p is upregulated in thymoma fluorescent reporter prove that the expression of FOXP3gene was modulated bymiR-125a-5p.Conclusion:1. These findings suggest that miRNA expression profiles get more vary inmyasthenia gravis patients with thymus near thymoma.2. And that miR-125a-5p might relate to the pathogenesis of myasthenia gravisby modulating FOXP3expression in myasthenia gravis patients withthymus near thymoma. |
