Part OneAnalysis on the infections status of saliva Helicobacter pylori in dental clinic patients in Lanzhou regionObjective:To determine saliva-prevalence of Helicobacter pylori (Hp) in Lanzhou, and then carry out healthy knowledge education about Hp related diseases.Methods:Helicobacter pylori were detected with bacterial culture, Gram stain microscopy urease test from saliva samples collected in941Lanzhou residents, Statistical analysis was performed with the chi-square test.Results:Saliva Hp positive rates of Lanzhou were42.72%, The infections status of Helicobacter pylori in different human groups existed significant difference.the female’s were47.89%, greater than38.44%of the male’s (p=0.004, x2=8.492); The highest positive rate was in the age group of over70; According to the living environment:The town’s were33.99%, less than50.93%of the villages’(p=0.000, x2=27.551),with no significant difference among which growthConclusions:The positive rate of Hp were associated with oral hygiene, oral disease, sex, age, and living environment. Part Two The selection of helicobacter pylori molecular biology method and oral helicobacter pylori detectionObjective:Choose molecular biology methods to detect H. pylori, which is more sensitive and efficient than conventional methods. And applied it to the detection of oral Helicobacter pylori infectionMethod:Helicobacter pylori were detected with PCR, SYBR Green real-time fluorescence quantitative PCR from gastric juice samples collected in32patients in gansu province people’s hospital gastroscope room. Choose the high sensitivity and specificity method to detect oral Helicobacter pylori. Helicobacter pylori were detected with SYBR Green real-time fluorescence quantitative PCR from plaque samples collected in138periodontitis patients in gansu province people’s hospital oral medicine, and analysis the correlation of plaque Hp between periodontitis and digestive tract diseases.Results:SYBR Green real-time PCR that we have established specificity is better, other strains stored in laboratory were no amplification.32cases of gastric juice samples for testing, the results showed that SYBR Green real-time fluorescence quantitative PCR detect Hp positive rate was90.63%, the qualitative PCR was only75.00%.So we choose the high sensitivity of SYBR Green real-time fluorescence quantitative PCR to detect oral Hp. the results showed that138cases of periodontitis plaque samples,the periodontal health group plaque Hp positive rate was30%, the periodontal disease group was74.69%, the periodontal disease with stomach disease group was92%, the difference was statistically significant.And serious, Hp positive rate increased with the degree of periodontal lesions.Conclusions:SYBR Green real-time fluorescence quantitative PCR can successfully detect gastric juniceand oral Helicobacter pylori with high sensitivity and strong specificity.The relation between oral Helicobacter pylori and occurrence of periodontitis development, digestive tract diseases are closely.
