| Objective To investigate the protective effects of Picroside II on renal ischemia-reperfusion injury in rats.Methods Forty healthy male Wistar rats were randomly divided five groups:â‘ ham-operated group;â‘¡I/R group:establish renal ischemia/reperfusion model of rats;â‘¢low-dose Picroside â…¡ group;â‘£middle-dose Picroside â…¡ group;⑤high-dose Picroside â…¡ group; At twenty minutes before surgery, Picroside â…¡ were administered by intrapertonel injection at dose of5mg/kg,10mg/kg,20mg/kg in the latter three groups. All the rats were sacrificed at24h after reperfusion.Serum levels of creatinine (Cr) and urea nitrogen(BUN) were determined, the GSH content and MDA content of renal tissue were deteced,the renal pathological alterations were observed by HE staining, the expression of TRAF6and caspase-3were deteced by immunohistochemistry technique.Result In I/R group, tubular epithelial cells were significantly impaired,but the pathological changes of glomerulus were not obvious.Compared with sham-operated group,the content of MDAã€TRAF6ã€caspase-3and GSH in ischemia-reperfusion group had statistical significance (P<0.05).Compared with ischemia-reperfusion group,the content of MDAã€TRAF6ã€caspase-3and GSH in Picroside â…¡ group had statistical significance(P<0.05).More over,compared with low-dose treatment group of Picroside â…¡,the content of MDAã€TRAF6and TRAF6in the middle-dose treatment and high-dose treatment of Picroside â…¡ had statistical significance(P<0.05).Conclusion Picroside â…¡ may provide beneficial actions by reducing oxidative stress,which can reduce apoptosis by down-regulating TRAF6.The protective effection has connect with the dose. |
