Objective:Through the inhibition of COX-2expression by siRNA, we want to observe the results of these genes (Bcl-2, Bax and Survivin) expression in MG-63.Methods:First of all, we will take the cells of log phase as a sample, which come from osteosarcoma MG-63in vitro subculture and design a siRNA which can interference specially COX-2gene expression. Secondly, we will use COX2-siRNAto transfect osteosarcoma MG-63under the mediate of lipidosome. Finally, in order to test the expression change of mRNA and protein of Bcl-2, Bax and Survivin after transfection, we will depend on two methods as RT-PCR and Western blotting.Results:With the result of the test mentioned above by RT-PCR and Western blotting, we have found that the special COX-2-siRNA can effectively inhibit COX-2expression. Besides that, we have also found this result can down-regulate the expression of anti-apoptosis protein Bcl-2and Survivin (P<0.05), however, up-regulate the expression of Bax(P<0.05).Conclusion:In this study, we discover a new research direction for treating osteosarcoma that is the inhibition of COX-2expression by siRNA, which can induce cell apoptosis and inhibit cell multiplication.
