| Objective To observe the effect of curcumin on human malignant melanoma cellline A375apoptosis and anti-cancer mechanism of action of curcumin. Methods Thehuman malignant melanoma cell line A375was divided into four groups, which wasrespectively treated by0,5.00,10.00,20.00μmol/L of different concentrations ofcurcumin for48h. Determination of cell proliferation by MTT assay; Cell cycle wasdetected by flow cytometry; Realtime PCR was used to detect the cell BIRC7mRNAexpression;the cell BIRC7protein and caspase-3protein were detected by Western blot.Result1.Curcumin can suppress cell proliferation and promote its apoptosis2.Curcumin can reduce the human malignant melanoma cell line A375BIRC7mRNAexpression: BIRC7mRNA expression relative ratio was (6.15±1.11),(4.54±1.23),(3.36±0.66) and (2.54±0.65) by0μmol/L,5.00μmol/L,10.00μmol/Lã€,20.00μmol/Lof different concentrations of curcumin treated respectively.3. Curcumin can downBIRC7protein expression: after treatment with0μmol/L,5.00μmol/L,10.00μmol/L,20.00μmol/L concentration of curcumin, the relative ratio of BIRC7protein expressionwas respectively (0.88±0.36),(0.71±0.28),(0.56±0.14) and (0.43±0.15). F=3.36,P=0.037<0.05.Conclusion Curcumin can promote human malignant melanoma cellline A375apoptosis.One of the mechanisms may be through downing BIRC7geneexpression. |
