| Objective The effect of Oncolytic adenovirus on different bladder cancer cells are different, In this study want to find out proteins’factors which may lead to this difference by Two-dimensional gel electrophoresis, the result can provide theoretical basis for further research on how to improve Oncolytic adenovirus’effect tothe bladder cancer. Methods (1) Detection the apoptosis rate of bladder cancer cells EJ, T-24and BIU-87which effected on adenovirus Ad-PSCAE-UP Ⅱ-E1A-AR72hours by flow cytometry assay, then compared their differences.(2) Collect bladder cancer cell EJ, T-24andBIU-87, extract the total proteins of the cells, Tissues from3EJ,3BIU-87and3T-24were analyzed by two-dimensional gel electrophoresi, Coomassie brilliant blue, used GS-800scan images, PDQuest7.0software analyze image to find out The differently expressed proteins, and then the identified protein by mass spectrometry detected. Results Adenovirus Ad-PSCAE-UP Ⅱ-E1A-AR function in cells after72hours, EJ cell apoptosis rate> T-24> BIU-87. we success get perfect repeatability gel image By two-dimensional gel electrophoresis. Through the analysis of the matching software on the gel, and find out the differences between the three groups of gel protein, after mass spectrum identification, these differences in protein in EJ, BIU-87, T-24expression quantity increasing is:calciumin, Actin, cytoplasmic1, Serine/arginine-rich splicing factor1, Heterogeneous nuclear ribonucleoproteins A2/B1, Endoplasmic reticulum resident protein29, Coiled-coil domain-containing protein153, glutamate dehydrogenase1, Peptidyl-prolyl cis-trans isomerase A, Galectin-1, Heterogeneous nuclear ribonucleoprotein L, Peroxiredoxin-4and Histone H2B type1-C/E/F/G/I.In EJ, BIU-87, T-24in the identified protein expression in turn reduce the amount of points is:Cytosol aminopeptidase and Mitotic checkpoint protein BUB3.Conclusion Adenovirus Ad-PSCAE-UP Ⅱ-E1A-AR can kill bladder cancer cell EJ, BIU-87, T-24. We find out the proteins’factors which may lead to this difference by Two-dimensional gel electrophoresis successful. |
