| PurposeCorneal ulcer was based on cornea tissues massive resolution induced byMMPs.This research focus on the inhibition effect of RAR r agonist CD437incorneal collagen degradation induced by IL-β.MethodsRabbit corneal fibroblasts were cultured in three-dimensional collagengels with Serum free medium, type I collagen,5×DMEM on ice until collagensolidification in incubator.Different concentration of CD437and plasminogenwere put on the surface of collagen,Collection of the culture supernatant,TheHYP amount was meassured.The expression and activity of MMP1,2,3,9wereevaluated by immunoblot analysis and gelatin zymography.ResultsCD437inhibit corneal collagen degradation induced by IL-1β in aconcentration and time-dependent manner.MMP1,2,3,9expression andactivation exposed to IL-1β were also inhibited by CD437.ConclusionsAs selective RAR r agonist,CD437can inhibit IL-1β–stimulated conealcollagen degradaation thereby be potent in the therapy of corneal ulcer. |
