Objective: By inducing myocardial injury of the rat with isoprenaline(ISO), this paper attempts to conduct analysis and research on the protectionrole potassium channel opener plays for myocardial cell. On this basic, theprotection effect of diazoxide on injured myocardial cell will be studied.Methods:30male SD rats (220to250g) shall be selected and divided into3groups randomly, including10for the normal control group,10for the ISOgroup and10for the DZ group. Each rat has to take the intraperitoneal injectionof5mg/kg ISO for3days continuously with once a day. After24hours of ISOinjection for the last time, pentobarbital sodium (60mg/kg) is used toanesthetize the rats with the collection of serum and heart tissue. Animal modelof ISO+DZ group preparation: DZ (5mg/kg) shall be provided30minutesbefore the ISO injection. As for the normal control group, the same dose ofnormal saline shall be injected to the peritoneal cavity of the rats.3days afterthe medical use, the myocardium of rats from each group shall be drawnrandomly to detect the lactic dehydrogenase (LDH) and creatine kinase (CK)which are seen as the markers of serum enzymology, the HE staining, and theexpression of HIF in the myocardial cell of rats by PCR detection.Statistical analysisStatistical analysis is carried out with the application of SPSS17.0system. The measurement data is represented by x±S, t test is adopted forcomparison within groups, and P <0.05indicates there is statisticalsignificance for the difference.Results:1. Histopathological detection for myocardiumThe result of HE staining: As for the normal control group, the fiber ofmyocardial cell is in alignment with clear cross grain and obvious cell nucleus,and there is no swelling of cell. There is necrocytosis for most of themyocardial cell in ISO group, and the myocardial fiber is in disorder withproliferation of fibrous tissue, accumulation and dissolving of cell nucleus, andinfiltration of a large amount of leukocyte. In terms of the ISO+DZ group, partof the myocardial cell is dead and falls off, and its cellular swelling is not asserious as that in ISO group.2. Inspection of serum enzymology Compared with the control group, theLDH and CK of ISO group is significantly higher, the difference of which iswith statistical significance (p <0.05); The LDH and CK of DZ group is lowerthan that in the ISO group, the difference of which is with statisticalsignificance (p <0.05).3. The HIF expression of the myocardium of rats The mRNA expression ofHIF in DZ group is obviously higher than that in ISO group, both of which arehigher than that in the control group.Conclusion: As is indicated by this experiment, diazoxide has the functionof protecting the injured myocardium, which is realized through mitoKATPopening.
