The Expression And Clinical Significance Of CXCR4、CXCR7in Esophageal Squamous Cell Carcinoma

Esophageal cancer is a common malignant tumor of upper digestive tractcancer.In the world, it is the eighth malignant tumor of incidence, the sixthmalignant tumor of mortality. It is a serious threat to the mankind. Fordecades,in China,the incidence and morbidity of esophageal cancerare ranked first in the world, prevention and treatment of esophageal cancer isfacing a severe test.Objective: In recent years,there are a lot of researches of chemokinein many kinds of tumor, suchasthe factorCXCL12and its receptores CXCR4andCXCR7, Recently,some researches from domestic and foreign, shownthe expression of the factor CXCL12and its receptores CXCR4, CXCR7had been found in many kinds of tumortissue, especially in mang kinds ofmalignant tumor tissues, They are closely related to the fucntion oftumortissue,such as proliferation, metastasis, angiogenesis this experimentaims to confirm the expression of receptor CXCR4, CXCR7inesophageal squamous cell carcinoma and its clinical significance.Methods:120samples of120patients,treated in Department ofThoracic Surgery in the Fourth Clinical Hospital of Hebei Medical Universityfrom January2012to Augest2013,were selected.The inclusion criteria were as follows: confirmed by surgical resection；no chemotherapy before surgery; postoperative pathologic results wereconfirmed, Another cases of adjacent normal Esophageal tissue samples(5cm above the edge of the cancerous tissue) were as the control group.Gender,age,degree of differentiation, lymph node metastasis,TNM stage, andother clinical data of the patients were recorded. Using the method ofImmuno-histochemical S-P was used to detect the expression levels ofCXCR4and CXCR7in the experimental group and the control group specimens. SPSS13.0statistical software was used to analysis the date in thisstudy, Using the chi-square test the relationship between the expression ofCXCR4and CXCR7with various clinical characteristics of esophagealcancer. The correlation of CXCR4and CXCR7by using Spearman rankcorrelation analysis. P<0.05was considered statistically significant.Result:1The positive expression of CXCR4in esophageal squamous cellcarcinoma was66.7%(Fig3); There was no expression in normal esophagealepithelium tissue(Fig1), the difference between the two results was statisticallysignificant (P=0.000),,the expression of CXCR4has nothing to do withpatients`age (P=0.658), gender (P=0.223), tumor size (P=0.824), the depthof invasion (P=0.327), the degree of tumor differentiation (P=0.275), but hascorrelated with the TNM stage(P=0.000) and lymph node metastasis(P=0.000).(Table1)2The positive expression of CXCR7in esophageal squamous cellcarcinoma was62.5%(Fig4);5.8%in normal esophageal epithelium tissue, thedifference was statistically significant (P=0.000),the expression of CXCR7hasnothing to do with patients`age (P=0.662), gender (P=0.134), tumorsize (P=0.434), the depth of invasion (P=0.145), the degree of tumordifferentiation (P=0.355), but has correlated with the TNM stage(P=0.000)and lymph node metastasis (P=0.000).(Table1)3In120cancer tissues,the rate of both positive expression of CXCR4and CXCR7was45.8%;the rate of CXCR4-positive and CXCR7-negativewas20.8%; the rate of CXCR4-negative and CXCR7-positive was13.4%;the rate of both negative expression of CXCR4and CXCR7was20%;Aboveall differences of the four combinations of CXCR4and CXCR7did notcorrelate with patients`age (P=0.456), tumor size (P=0.580), depth of tumorinvasion (P=0.168), but correlated with patients`gender (P=0.031), tumordifferentiation (P=0.037), TNM staging (P=0.000), number of lymph nodemetastasis (P=0.000).(Table2) 4The expression of CXCR4had correlate with the expression of CXCR7(P=0.000)(Table3).Conclusion:1The expression rates of CXCR4and CXCR7in esophageal squamouscell carcinoma compared to normal esophageal epithelium (from the tumormargin>5cm) was significantly higher, and their expressions do not correlatewith age、gender、the depth of invasion、the tumor size and the degree of tumordifferentiation,but associated with the TNM-stage and the number of lymphnode metastasis.2The expression of CXCR4associated with the expression of CXCR7,both of them have a synergistic effect, the number of the CXCR4-positive andCXCR7-positive is more men than women.
