| Objective:To observe the effect of HUQIAN PILL on bone mineral destiny (BMD) and transforming growth factor-beta2(TGF-β2) in right side of the proximal femur of ovariectomized rats, then the correlation between TGF-β2and BMD was analyzed. To investigate the effect of HUQIAN PILL on BMD and TGF-β2in ovariectomized rats.Methods:Forty female Sprague-Dawley rats were randomly divided into four groups:the HUQIAN PILL group, the estrogen group, the ovx group and the sham-operation group. Each group had10rats. Rats in sham-operation group had their abdominal cavity opened and a piece of ovary surrounding fatty tissue excised before closing the abdomen. The osteoporosis model were established by bilateral ovariectomy in other groups. Each group began treatment4weeks after operation. The HUQIAN PILL group were given the water decoction of HUQIAN PILL. The estrogen groups were given the water decoction of estrogen medicine. The other groups were given the distilled water. Each group began treatment once a day for12weeks. To use of dual-energy X-ray absorptiometry of the femur BMD measured in rats, which had proved model was succeed. After12weeks, the histopathological response changes of right side of the proximal femur of rats in each group were compared, then TGF-β2on osteoblast cells were detected by immunohistochemical method.Results:One rat died unexpectedly in the HUQIAN PILL group and the estrogen group during the study of the40rats. Comparison BMD in right side of the proximal femur of ovariectomized rats, between the model group and the sham-operation group on4weeks after operation, which all significantly decreased (P<0.05), indicating that models were successfully established. After12weeks, observation of the proximal femur (right side) pathological slices with light microscope, the HUQIAN PILL group better than the other groups. Comparison of BMD of the proximal femur (right side) of rats in each group:the HUQIAN PILL group, the estrogen group and the sham-operation group were higher than in ovx group at different degree (P<0.05). No significant difference between the HUQIAN PILL group and the estrogen group and the sham operation group (P>0.05). No significant difference between the estrogen group and the sham operation group (P>0.05). Comparison in the expression of TGF-(32of rats in each group:the HUQIAN PILL group, the estrogen group and the sham-operation group were higher than in ovx group at different degree (P<0.05). The statistically significant difference between the HUQIAN PILL group and the sham operation group (P<0.05). The statistically significant difference between the estrogen group and the sham operation group (P<0.05). No significant difference between the estrogen group and the HUQIAN PILL group (P>0.05). Then the correlation between TGF-β2and BMD was analyzed. There was a positive linear correlation between TGF-β2and BMD.Conclusion:The osteoporosis symptoms of patients were obviously improved by treatment of the HUQIAN PILL. Then the active bone cell differentiation, proliferation, increased bone trabecula, and then got close to normal or nearly normal levels. It can increase in the expression of BMD and TGF-β2by treatment of the HUQIAN PILL. There was a positive linear correlation between TGF-β2and BMD, which can proved correlation analysis between them. Then explain the intervention the right side of the proximal femur BMD change will affect osteoblasts in the expression of TGF-β2. |
