The main chemical components of Jasminum officinale L.var. grandiflorum iridoid glycosides, flavonoids and triterpenoid saponins compounds, iridoid glycosides mainly oleuropein.In recent years, with domestic and foreign research scholars oleuropein deepening and found that it has many pharmacological activities, causing widespread concern.Oleuropein with anti-VHSV, HIV, HBV and other viruses in the role; anti-tumor effect; antioxidant; hypoglycemic effect; on the cardiovascular system; on Metabolism; antibacterial activity and a series of anti-inflammatory effects Pharmacological activity.J. officinale of total iridoid glycosides as the main active ingredient, the total iridoid glycosides mainly oleuropein, oleuropein is a phenol crack iridoid glycoside, oleuropein soluble in ethanol, acetone, Pyridine, etc., can be dissolved in water, alcohol, ethyl acetate, almost insoluble in ether, petroleum ether, chloroform and so on.So far China has not yet SuXinHua on the total iridoid glycoside concentration purification process optimization reported in the literature.Therefore, this study uses orthogonal J. officinale on the total iridoid glycosides were optimized extraction process and to develop quality standards, then the polyamide resin as the stationary phase, select the appropriate mobile phase, applied pressure column chromatography, J. officinale herbs total iridoid glycosides in oleuropein to explore a rapid extraction, separation and purification of the active ingredient of SuXinHua simple and practical medicine and ultimately to determine the best method of extraction and purification process, and setting the total Iridoid glycosides and determination of oleuropein methods and quality standards. J. officinale for the further development and utilization of the total iridoid glycoside of the biological activity and provide experimental basis for the pharmacological activity after experimental basis.The first part J. officinale total iridoid glycosides Extraction TechnologyObjective:To investigate the optimal extraction process for total iridoid glycosides from J. officinale.Methods:In this research, the content of total iridoid glycosides was measured by the spectrometer, and then the calculated yield of total iridoid glycodides(based on medicinal materials) was taken as evaluation index. Finally, the L9(34) orthogonal test table was adopted to evaluate the effect of different extraction technologies:cold immersion with water, decoction with water, immersion with cold ethanol and reflux with ethanol.Result:Optimal conditions for the water cold soak A3B3C3, which uses12times the amount of extracted distilled water cold soak3times per72h; Optimal conditions of water boiling A3B1C3, which uses16times the amount of boiling distilled water3times, each time1h; Optimum conditions for alcohol cold soak A2B1C3, which uses12times the amount of60%ethanol extract cold soak3times per48h; Optimum conditions for alcohol reflux A3B3C1, which uses12times the amount of60%ethanol extract3times, each time2h. The optimal combination of the various methods of total iridoid glycosides yield little difference.Conclusion:12times with a cold soak of60%ethanol extracted3times (each48hours) of the extraction method can obtain high extraction rates, and stable and feasible. Total iridoid glycosides standard curve: A=0.0289C-0.0049(R2=0.9998),3.96-19.80μg/ml inside the scope of sex is good, The average recovery value is102.64%, RSD=0.65%(n=6). Oleuropein standard curve: Y=133959x+23.44(R2=0.9998)0.02-0.10mg/ml inside the scope of sex is good, The average recovery value is100.12%, RSD=0.883%(n=6).Conclusion:The method is stable, accurate, high precision, specific, simple feasible, for optimization of extraction and purification process of the content.
