Objective: To investigate the mechanism of the miR-124inhibiting the coloncancer cells growth and invasion.Methods: Cell growth was evaluated by MTT. Cell invasion potential wasevaluated by scratch test and transwell assay. miR-124targeting Vimentin wasevaluated by luciferase reporter vector. Immunocytochemistry was used to detected theexpressions of molecules with invasion.Results: miR-124could inhibited growth and proliferation of SW620cells in atime-dependent manner. The OD values were0.287±0.010,0.353±0.006and0.376±0.007after tansfecting miR-124mimics for24,48and72h by MTT. The ODvalues were0.436±0.014,0.551±0.015,0.642±0.021after tansfecting miRNAnegative control for24,48and72h by MTT. It’s certified that miR-124could inhibitcolon cancer cell motility and invasion identified by scratch test and transwell assay.Vimentin was a target gene of miR-124which evaluated by luciferase reporter vector.miR-124downregulated the expression of Vimentin and MMP-2protein byImmunocytochemistry and Western blot.Conclusions:1. miR-124inhibits the colon cancer cells growth and invasion.2. Vimentin is the target gene of miR-124.3. miR-124can inhibit colon cancer cell growth and ivasion by inhibiting theexpression Vimentin and MMP-2.
