We collected210cases clinic examination healthy individuals as the control group and200cases treated in our hospital as patients group with Type2diabetes mellitus (T2DM) from the northern Han Chinese population. White blood cells and serum were separated, leukocyte genomic DNA were extracted using SDS-proteinase K-phenol/chloroform method, ApoE genotype was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) levels were tested with the automatic biochemical analyzer. This study intends to reveal the ApoE gene polymorphism of T2DM and the plasma lipid.We found that ApoE ε3/3genotype most frequently, ε3/4genotype and ε2/3genotype frequency followed, whereas ε2/2, s2/4and s4/4genotype frequency is very low in the case group and the control group. The ε3/4genotype frequency in diabetes group was higher than that in control group. The levels of serum TC, TG and LDL-C in diabetes group with ε3/4were higher than those in control group, while the levels of HDL-C in diabetes group with ε3/4were lower than those in control group. Metabolic disorders were more serious in diabetes group with s3/4than the other genotypes.The ApoE ε3/4genotype was related to the T2DM and abnormal lipid metabolism. And further reveals the geographical specificity of ApoE gene polymorphisms, genetic data supplemented with regional characteristics in China, and provide valuable basic data for the overall study the structure and function of ApoE gene.
