| Key words: Breast carcinoma; Microvessle density; Vascular endothelial growthfactor; basic-Fibroblast growth factor; CorrelationObjectiveTo study the expression of microvascular density (MVD) in breast carcinomatissue, vascular endothelial growth factor(VEGF), basic fibroblast growth factor (bFGF)in tissue and serum in breast carcinoma, to explore the relationship between MVD,VEGF, bFGF and biology behavior of breast carcinoma. The number of tumormicrovascular was calculated through the expression of CD34, to study the relationbetween MVD and VEGF, bFGF, and to discuss the role of these indicators in theoccurrence, development, invasion and metastasis of breast carcinoma.MethodsThe expression of VEGF, bFGF and CD34in the tissue of breast carcinoma andbreast fibroadenoma were detected by using immunohistochemical method, thecorrelation between three indicators above and pathological features such as age,clinical stage, tumor diameter, pathological types and lymph node metastasis wereanalyzed. Correlation analysis was conducted to analyze the relationship between MVDin breast carcinoma tissue and VEGF, bFGF in serum and tissue.Results1. The positive expression rate of VEGF, bFGF protein in breast carcinoma tissuewas71.3%,68.8%respectively, which was significantly higher than20.0%,25.0%ofbreast fibroadenoma(χ2=21.09,12.76, P<0.05). 2. The count of MVD in breast carsinoma and breast fibroadenom were33.81±10.95,17.41±6.04, it was statistically significant (t=6.44, P<0.05).3. The positive expression rate of VEGF increased as the increase of tumordiameter, TNM clinical stage, lymph node metastasis (P<0.05), but nothing on age,pathological type (P<0.05). The positive expression rate of bFGF increased as theincrease of TNM clinical stage, lymph node metastasis (P<0.05), but nothing on age,tumor diameter, pathological type (P>0.05).4. The count of MVD in stage of â…¢, â…£ was significant higher than that stageofâ… ,â…¡, and MVD expression was more frequently found in tumors with lymph nodemetastasis than in those without (P<0.05).5. The expression of serum VEGF, bFGF in breast carcinoma was remarkablyhigher than that of healthy control (P<0.05).6. With the increase of diameter increased and TNM clinical stage, the expressionlevel of serum VEGFã€bFGF increased, and serum VEGF〠bFGF expressed more intumors with lymph node metastasis than in those without (P<0.05), but nothing on age,pathological type (P>0.05).7. The expression level of VEGF in tissue of breast carcinoma was positivecorrelated with bFGF(r=0.685, P<0.05), and they had the same correlation in serum inbreast carcinoma(06, P<0.05).8. The count of MVD in tissue of breast carcinoma was positive correlated withVEGF, bFGF(r=0.594,0.525,P<0.05), and they had the same correlation in serum inbreast carcinoma(r=0.472,0.507, P<0.05).Conclusion1. The expression of VEGF, bFGF in serum and tissue, and MVD in breastcarcinoma are higher than those in mammary gland tumor and healthy controls.2. The expression of VEGF, bFGF in serum and tissue, and MVD in breastcarcinoma are correlated with pathological characteristics such as diameter, TNMclinical stage, lymph node metastasis, detection of the expression of bFGF, VEGF andMVD has important reference value in the judge of malignant degree and prognosis of breast carcinoma3. There is highly positive correlation between VEGF and bFGF in serum andtissue in breast carcinoma, and the same relation was found between VEGF, b FGF andMVD. They are involved in the process of the development and metastasis of breastcarcinoma, and may be important targets for anti-angiogenesis therapy in patients withbreast carcinoma. |
