Due to the high incidence, morbidity and mortality, cerebral ischemia disease brings great pain and burden to society and family, but the treatment is very limited. Classical Tibetan prescription, Tibetan prescription A, Tibetan prescription B, Tibetan prescription C often used in the treatment of ischemic diseases and have good effect, and have received deeply recognition by the Tibetan people. The purpose of this experiment is to investigate the efficacy of the combined using of these three kinds of prescription, and explore the rationality of its dosing method.Methonds:(1)Study on the protective efficacy of the combined using of these three kinds of prescription:The Sprague-Dawley rats were randomized and averagely divided into seven groups.Sham operation and pMCAO model, normal saline was given orally ever)’ day(1mL/100g·bw); pMCAO+Nimodipine, nimodipine was given orally ervery day(1.89mg/100g·bw); pMCAO+Tibetan prescription A, Tibetan prescription A was given orally ervery day(21mg/100g·bw): pMCAO+Tibetan prescription B, Tibetan prescription B was given orally ervery day(94.5mg/100g·bw); pMCAO+Tibetan prescription C. Tibetan prescription C was given orally ervery day(147mg/100g·bw); pMCAO+drug combination, Tibetan prescription A(*mg/100g·bw), Tibetan prescription B(*mg/100g·bw), Tibetan prescription C(*mg/100g·bw)were separately given ervey day. Establish inproved pMCAO model after7day’s intragastrical administration, and determine the cerebral infarct size(TTC method); Behavioral symptoms(zea Longa method); water content in brain(drying and weighing); the activities of SOD in serum(xanthine oxidase method): the activities of LDH in serum(pyruvate method); the content of MDA in serum(TBA method).(2)Study on the therapeutic efficacy of the combined using of these three kinds of prescription: The Sprague-Dawley rats were randomized and averagely divided into seven groups, and implemented pMCAO surgery. After surgery, Sham operation and pMCAO model, normal saline was given orally every day(1mL/100g·bw); pMCAO+Nimodipine, nimodipine was given orally ervery day(1.89mg/100g·bw); pMCAO+Tibetan prescription A, Tibetan prescription A was given orally ervery day(21mg/100g·bw); pMCAO+Tibetan prescription B, Tibetan prescription B was given orally ervery day(94.5mg/100g·bw); pMCAO+Tibetan prescription C, Tibetan prescription C was given orally ervery day(147mg/100g·bw); pMCAO+drug combination, Tibetan prescription A(*mg/100g·bw), Tibetan prescription B(*mg/100g·bw), Tibetan prescription C(*mg/100g·bw)were separately given ervey day.5days after, determine the cerebral infarct size(TTC method); water content in brain(drying and weighing); the activities of SOD in serum(xanthine oxidase method); the activities of LDH in serum(pyruvate method); the content of MDA in serum(TBA method); HE stain; Nissl’s staining(toluidine blue method); Immunohisto-chemistry(S-P method).Results:(1)Study on the protective efficacy of the combined using of these three kinds of prescription:Compared with the pMCAO model group, the combined using of the three Tibetan classical prescription can reduce the cerebral infarct size, water content in brain issue, increase the activity of SOD in serum, and reduce the content of MDA in serum.(2)Study on the therapeutic efficacy of the combined using of these three kinds of prescription:Compared with the pMCAO model group, the combined using of the three Tibetan classical prescription can reduce the cerebral infarct size, increase the activity of SOD in serum, reduce the activity of LDH in serum, reduce the content of MDA in serum, effectively reduce degree of pathological changes and damage to the nerve cells in the brain tissue, promote the expression of Bcl-2protein, reduce the expression of Bax, Caspase3, CytC, HIF-la, NFKBp65, VEGF protein after cerebral ischemia injury.Conclusions:Preliminary experimental results show that the conmbined using of these three classical Tibetan prescription can relieve the damage of brain tissue in cerebral ischemia disease, and have certain protective and therapeutic effect on ischemia disease. Compared with single dose, the conmbined using of Tibetan prescription can improve the body’s antioxidant ability, reduce the degree of cerebral edema, reduce the apoptosis of cells.
