Objective:The purpose of this study was to explore the anthocyanins’antioxidant activity from purple sweet potato, anti-aging effects and the protective effect of alcohol-induced liver injury and provide theoretical support for the further development and utilization of purple sweet potato.Method:Clear vitro assay purple sweet potato anthocyanins DPPH· ability to affect the liver homogenates glutathione peroxidase (GSH-Px) activity, as well as H2O2-induced mouse liver homogenates MDA generation and inhibition of erythrocyte hemolysis.Preparation of the use of D-galactose aging model mice, after a purple sweet potato anthocyanins150,300,600mg/kg administered orally, through general observation and organ index detection and measurement of liver and brain tissue homogenates uniform pulp SOD, MAO, MDA levels and other changes.Through the establishment of alcoholic liver injury model, prophylactic administration by detection of serum ALT, MDA content, GSH-Px activity alter the protective effect of AST activity in liver homogenates and purple sweet potato anthocyanins study of alcoholic liver injury.Result:Purple sweet potato anthocyanins on DPPH· scavenging IC50of approximately2.2μg/ml, vibrant purple sweet potato anthocyanins value added group GSH-Px is greater than the control group, MDA generated significantly less amount of purple sweet potato anthocyanins group in the control group, purple sweet potato anthocyanin concentration of2.0mg/L, erythrocyte hemolysis inhibition rate was57.4%.Old experiment, mice exhibited significantly reduced activity, fur gloss decrease hair loss and other signs of aging, brain MAO activity increased, brain and liver tissue SOD activity decreased, MDA content increased. Compared with the model group, the activity of brain MAO purple sweet potato anthocyanins dose group reduced significantly increased brain and liver tissue SOD activity and reduce MDA content.Alcoholic liver injury experiments, compared with the control group, model group AST, ALT activity increased, rising liver MDA, GSH-PX activity decreased; dose group AST and ALT activity decreased, MDA content in the liver decreased, GSH-PX activity enhanced. Conclusion:Purple sweet potato anthocyanins can effectively remove DPPH·, enhancing the activity of GSH-Px in liver homogenates, could significantly inhibit the formation of H2O2-induced MDA of mouse liver homogenates and hemolysis of red blood cells.Purple sweet potato anthocyanins had obvious anti-aging effects on subacute aging model induced by D-galactose and this anti-aging effects were may be related to its antioxidant effects. While giving the mice of purple sweet potato anthocyanins and alcohol can effectively reduce the damage of alcohol on the liver of mice, and there is a certain dose-effect relationship, and this protective effects were also may be related to the antioxidant effects of purple sweet potato anthocyanins.
