Objective: To investigate the effects of lipoxin A4(LXA4) oninflammatory related factors induced by uric acid (UA) in human umbilicalvein endothelial cells (HUVECs).Methods: HUVECs were treated with LXA4(5,25and50ng/mL) invarious concentrations prior to UA(12mg/dL) exposure. Tumor necrosisfactor-alpha(TNF-α), interleukin-1β(IL-1β) and interleukin-6(IL-6) wereanalyzed with ELISA, real-time quantitative reverse transcription polymerasechain reaction (RT-qPCR). The effects of LXA4on UA induced p38MAPKand NF-κB/p65were observed with western blot.Results: Stimulation of HUVECs with12mg/dL UA markedly increasedTNF-α, IL-1β and IL-6production(P<0.01). Pretreatment with LXA4significantly inhibited UA-induced production of TNF-α, IL-1β and IL-6in aconcentration dependent manner(P<0.01). The mRNA expression of TNF-α,IL-1β and IL-6in response to UA were also decreased by LXA4(P<0.05).Western blot analysis showed that the expression of phospho-p38andphosphor-NF-κB/p65was significantly increased in the UA group (P<0.05),but attenuated significantly in the50ng/mL LXA4prior to UA exposuregroup.Conclusion: LXA4may inhibit the expression of inflammatory relatedfactors at least in reducing p38MAPK and NF-κB/p65phosphorylation toplay a role in anti-inflammation.
