| Blood substitutes, mainly named artificial oxygen carriers, could carry oxygen and transport oxygen to tissues. They have no blood type antigen. no infection; stability for long-term storage at room temperature for stockpiling for any emergency; low toxicity and prompt metabolism; physicochemical properties that are adjustable to resemble those of human blood; reasonable production expense and cost performance. In this work, amphiphilic block copolymers containing different functional group have been synthesized from PEG and LA, and then could self-assemble to form polymer vesicles in aqueous solution. Hemoglobin could be contained into polymersome to prepare biodegradable hemoglobin nanoparticles, and they have the ability of carrying oxygen.(1)Amphiphilic block copolymers have been prepared from LA and PEG as macro initiators. The properties of the copolymers and their corresponding polymersome have been investigated through HNMR, GPC, DLS, TEM, and pyrene probe method.(2)The polymersome from the above-mentioned copolymers could self-assemble in aqueous solution, made use of lyophilized powder for reconstitution and film hydration method to encapsulation hemoglobin. With the help of DLS, TEM, UV, it has proved that hemoglobin successfully be contained into polymersome.(3)The various physicochemical properties of hemoglobin nanoparticles have been discussed and determined the content of hemoglobin, oxidized stability, and carrying oxygen properties of hemoglobin nanoparticles solution. In addition, we discussed that the effect on hemoglobin nanoparticles and the biocompatibility with whole blood. |
