Diabetes is a kind of common endocrine disorder diseases, and has become a serious epidemic non-infectious disease. Corn Stigma as corn dry grasses style and the column head, for our traditional Chinese medicine. Its source abundant, low cost, and it has not been comprehensive development and utilization for medicinal resources. But the majority of corn silk has not yet been fully utilized. That is because the effective part of corn silk for lowering blood glucose is not clearly related. If we can produce the pharmacological active ingredient research clearly, then will develop coin silk extracts hypoglycemic agents for doing preparation, and also lay a theoretical basis for coin silk resource development.This paper firstly studied polysaccharides, flavonoids, saponins which were extracted from corn stigma on alloxan diabetic mice blood glucose lowering effect, and had selected pharmacological effect of hypoglycemic effective site. Secondly, used microwave extraction and heating extraction methods to optimize the flavonoids extraction conditions to contrast the content of flavonoids under the optimum extraction conditions. Finally, determined the flavonoids contents from five kinds of corn in different periods of Liaoning Province. The results were as follows:1-. The corn stigma polysaccharides, flavonoids, saponins lowered the blood glucose of diabetic mice by pharmacological experiments. The flavonoids played the role of hypoglycemic effect more significantly. Initially concluded that flavonoids were the most effective ingredients of hypoglycemic extracts of corn stigma.2n The results of microwave extraction flavanones orthogonal design showed that:the optimum extraction conditions of flavonoids was ethanol concentration80%, solid-to-liquid ratio1:25, microwave power640w, microwave extraction time4minutes. Under the same conditions, the total flavonoids were2.43%. Under the optimal conditions of heating extraction flavanones orthogonal design, ethanol concentration80%, solid-to-liquid ratio1:20, extracting temperature70℃, extracting time1.5hours, the total flavonoids were1.07%. The contrast results illustrated microwave exraction was a efficient timesaver method. 3、The total flavonoids contents of milky stage of corn stigma was less than mature period. Contents of total flavonoids were obviously different in varieties of corn stigma and differ at most two times.
