| Objective To study the inhibitory effect on allergic asthma by adoptive transfer of CD8a+CDllc+and CD8α-CDllc+dendritic cells (DC) from Schistosoma japonicum-infected mice. Methods CD8α+CDllc+and CD8α-CDllc+DCs from schistosoma japonicum-infected mice and naive mice were respectively isolated and purified using CD8a+and CD11c+microbeads. Thirty BALB/c mice were divided into six groups randomly.5×105CD8α-CD11c+DCs of Schistosoma japonicum-infected mice were adoptively transferred to each mouse in Group A by tail vein, while5×105CD8α-CD11c+DCs of naive mice were adoptively transferred to each mouse in Group B in the same way,5×105CD8α+CD11c+DCs of Schistosoma japonicum-infected mice were adoptively transferred to each mouse in Group C by tail vein, while5×105CD8a+CD11c+DCs of naive mice were adoptively transferred to each mouse in Group D in the same way, Group E was asthma group, Group F was control group of naive mice.1hours later, mice in Group A, B, C, D and E were sensitized and challenged with OVA to induce asthma.4weeks later, all the mice were killed. Sera were collected for measurement of OVA-specific IgE; Bronchoalveolar lavage fluids (BALF) were collected for measurement of various cytokines (IL-4, IL-5, IL-10, IL-12and TGF-β); Spleen cells were collected and cultured,72h later, spleen culture supernatants(SPC) were collected also for determination of IL-4, IL-5, IL-10, IL-12and TGF-β, Lungs were collected for histological analysis. Results Compared with mice in Group B, C, D, and E pulmonary inflammation of mice in Group A diminished markedly (P<0.05) Moreover, the levels of IL-4in SPC of mice in Group Aã€Bã€Cã€Dã€E and F were (33.74±4.23) pg/ml,(77.63±5.11) pg/ml,(97.21±7.02) pg/ml,(75.50±22.37) pg/ml,(118.45±9.40) pg/ml and (1.74±1.76) pg/ml respectively; The levels of IL-5were (24.47±3.67) pg/ml,(133.76±31.85) pg/ml,(197.10±13.31) pg/ml,(167.10±15.07) pg/ml,(257.97±36.75) pg/ml and (0.96±0.77) pg/ml respectively; The levels of IL-10were (857.58±33.03) pg/ml,(254.16±26.28) pg/ml,(232.00±27.42) pg/ml,(176.44±6.15) pg/ml,(164.42±26.47) pg/ml and (142.23±11.96) pg/ml respectively. The levels of IL-12were(484.61±33.46)pg/ml, (271.68±30.34) pg/ml,(211.90±17.60) pg/ml,(235.63±24.58) pg/ml,(202.55±14.71) pg/ml and (96.55±13.51) pg/ml respectively. The levels of TGF-β were (2011.50±269.91) pg/ml,(1017.75±153.48) pg/ml,(937.13±92.52) pg/ml,(970.88±69.45)pg/ml,(834.63±48.94)pg/ml and(731.81±32.46)pg/ml respectively. Compared with mice in Group B, C, D and E, levels of IL-4ã€IL-5in SPC of mice in Group A decreased significantly (P<0.05), while levels of IL-10ã€IL-12ã€TGF-β in SPC of mice in Group A increased significantly (P<0.05). The levels of IL-4in BALF of mice in Group Aã€Bã€Cã€Dã€E and F were (25.85±9.00) pg/ml,(44.83±6.07) pg/ml,(74.24±8.28) pg/ml,(70.23±12.20) pg/ml,(79.15±8.74) pg/ml and (24.22±2.31) pg/ml respectively; The levels of IL-5were (25.34±1.85) pg/ml,(143.11±5.28)pg/ml,(140.17±10.05)pg/ml,(134.33±17.63)pg/ml,(149.22±0.90) pg/ml and (19.95±5.40) pg/ml respectively; The levels of IL-10were (369.59±46.72) pg/ml,(192.41±32.84) pg/ml,(168.92±13.64) pg/ml,(164.75±28.44) pg/ml,(144.54±10.19) pg/ml and (151.63±10.40) pg/ml respectively. The levels of IL-12were (755.68±30.08)pg/ml,(555.30±41.33)pg/ml,(577.05±23.69) pg/ml,(593.63±29.59) pg/ml,(468.97±13.05) pg/ml and (340.47±18.28) pg/ml respectively. The levels of TGF-β were (2589.38±411.52) pg/ml,(992.44±157.06) pg/ml,(1017.13±189.45) pg/ml,(1257.75±92.81) pg/ml,(654.63±127.52) pg/ml and (1104.63±275.12) pg/ml respectively. Compared with mice in Group B, C, D and E, the levels of IL-4and IL-5of mice in Group A decreased significantly (P<0.05), and the levels of IL-10,ã€IL-12ã€TGF-β were increased (P<0.05). The levels of OVA-specific IgE were (214.15±27.39) pg/ml,(987.77±35.58) pg/ml,(1000.85±56.64) pg/ml,(1035.85±22.70) pg/ml,(1118.63±33.62) pg/ml and (155.59±1.17) pg/ml. Compared with mice in Group B〠Cã€D and E, the levels of OVA-specific IgE of mice in Group A decreased significantly (P<0.05). Conclusion CD8α-CD11c+DC from S. japonicum infected mice can significantly inhibit allergic asthma. |
