Enterococcus Faecalis Effect In Expression Of TLR2in Experimental Rat Periapical Periodontitis

Objective:To study Enterococcus faecalis effect in the expression of Toll likereceptor2(TLR2)in the process of rat periapical periodontitis.Methods:40adult male Sprague-Dawley rats with the right side of the mandibularfirst molar after opening the chamber,were randomly divided into two groups (A、Bgroup),each group had20rats.Group A,medullary cavity is filled with PBS cottonballs,glass ionomer temporarily closed,enclosed in Enterococcus faecalis bacteriasuspension cotton ball in the medullary cavity of the B group,the glass ionomertemporarily closed; the mandibular left first molars were used for control.A、B groupsand then the first week, the second week,the third week,the forth week, four time pointsdivided into A1、A2、A3、A4、B1、B2、B3、B4eight subgroups,each subgroup withfive rats.At the specified time point,rats were sacrificed by ether,separation of themandible,dug a spoon scraping the apical region of the mandibular first molarperiodontal ligament,alveolar bone area,extracted tissue RNA,the reverse cDNA,theexpression of TLR2gene was used the Real-time PCR method for detection of eachgroup.The results were analysis by SPSS16.0.Results:1.TLR2was expressed in control.The expression of TLR2gene of A group,1weekto2week,was gradually increased and reached a peak at3week,4week began todecline,each group has statistically significant to control(P<0.05).Unless the differencewas not statistically significan between1week and4week(p>0.05), the differencesbetween other groups have statistically significant(P<0.05).2.The expression of TLR2gene of B group,1week to2week,was graduallyincreased and reached a peak at3week,4week began to decline,each group has statistically significant to control(P<0.05).Unless the difference was not statisticallysignifican between1week and4week(p>0.05), the differences between other groupshave statistically significant(P<0.05).3.The expression of TLR2gene amount of each time point the group B were higherthan the group A,the difference was statistically significant (P <0.05).Conclusion:1.Toll like receptor2involved in the whole process of periapical,the expression ofToll like receptor2was closely related to the periapical periodontitis, in which play animportant role.2.Toll like receptor2closely related with the process of the main bacteriaEnterococcus faecalis infected periapical periodontitis,Enterococcus faecalis enablesincreased expression of Toll like receptor2.
