Objectives:To evaulate the cytotoxicity and antibacterial effects of Silagum-Comfor silicone-based soft liner(SLC) varnish containing Ag/TiO2in vitro.Methods:Diameter10mm,2mm thick silicone-based liner circular specimens were madeand divided into6groups randomly. Ag/TiO2were incorporated into SLC varnishaccording to the weight rate of0%、0.5%、1.0%、1.5%、2.0%、2.5%agent/materialrespectively.Then the mixed varnish were polished the SLC specimens equably by theformer groups and then antibacterial specimens (ATSLC) were made. MTT assaymethod was used to evaluate the3T3cell cytotoxicity of ATSLC in vitro. TheMinimal Bactericidal concentration (MBC) of Ag/TiO2against C. albicans was testedby Liquid Dilution method. Surface roughness(Ra) measurements of ATSLC weretaken. The antibacterial effect to C.albicans of ATSLC was examined by thepellicle-sticking method.Results:1. The cytotoxicity were increasing gradually with the Ag/TiO2concentrationincreasing of ATSLC.Compared with the positive control group,the difference arestat-istically significant. The cytotoxicity of ATSLC were grade0-2.2. The Minimal Bactericidal concentration (MBC) of Ag/TiO2against C.albicans was0.156mg/mL.3.The Ra of ATSLC were increased with Ag/TiO2agent concentration increased,however the differences were not statistically significant compared with the controlgroup.4.With the increasing of Ag/TiO2antibacterial agent,the antibacterial rate of theATSLC increased too. When the addition amount reached to2.0wt%and2.5wt%,the antibacterial rate up to100%.Conclusions:1. The cytotoxicity of varnish containing Ag/TiO2to3T3cell were grade0-2and it was compliant with the safety standards of medical equipment.2. Varnish containing Ag/TiO2had excellent antibacterial effect and it canimproves the antibacterial effect of silicone-base soft liner greatly.3. The recommended incorporation ratio of Ag/TiO2in SLC varnish is2.0wt%.
