objective:By observing the Zhilonghuoxuetongyu capsule on focal cerebral ischemia in rats neovascularization and related factors, to explore the mechanism of focal cerebral ischemia in rats microvascular protection,From the perspective of promoting brain neovascularization,to explore the role of Zhilonghuoxuetongyu Capsule treatment of ischemic stroke neurological function recovery, to provide a new basis for its treatment of cerebral infarction.Methods:120healthy male adult SD rats, body weight (250-300) g were randomly divided into six groups (30spare):Sham operation group, model group,Zhilonghuoxuetongyu capsule high-dose group(referred to as "high-dose group), Zhilonghuoxuetongyu capsule middle dose group (referred to as the" middle dose group), Zhilonghuoxuetongyu capsulelow dose group (referred to as "low-dose group), Buchang Naoxintong the control group(referred to as Naoxintong group), eachgroup (n=20).Experimental to Zea longa’s method for standard production of middle cerebral artery occlusion (MCAO) model, only the sham-operated group median incision, but do not insert line bolt.Each treatment group were fed in the wake of the animal, sham group and model group were given an equal volume of saline, to score Zea longa’s ratings standard5-point scale neurological deficit scores, after7days, serum VEGF levels; perfusedfixed, the brains were removed, the HE staining morphological changes; immunohistochemistry was used to detect cerebral cortex ischemic area micro vessel density (CD31), TGF-betal, Ang-1, Tie-2protein expression.Results:(1) Rats and nerve function deficit score.Sham-operated rats coat gloss, agile, and normal feeding, responsive to around change, does not appear symptoms of neurological deficit;Model rats coat lose their luster, state of malaise, decrease in food intake and locomotor activity, apparent symptoms of neurological deficit, neurological deficit scores between2.00-3.96;High-dose rats coat gloss, good spirits, normal feeding, locomotor activity recovered rapidly, neurological deficit scores between1.41-2.47.Compared with the sham group, model group neurological deficit score was significantly higher, the difference was significant (P<0.01), the rats administered neurological deficit scores were higher, the difference was statistically significant (P <0.05).Compared with the model group, the high-dose group, middle dose group and Naoxintong group neurological deficit scores were significantly reduced, the difference was significant (P<0.01), low-dose group rats neurological deficit scores also have a certain degree oflower, the difference was statistically significant (P<0.05).Compared with the low-dose group, high dose group, middle dose group and Naoxintong group of neurological function deficit scores were lower, the difference was statistically significant (P <0.05).High-dose group, middle dose group,Naoxintong group neurological deficit score between the pairwise comparison, there was no significant difference (P>0.05).(2)Zhilonghuoxuetongyu capsule on cerebral ischemia in rat serum VEGF levels:Compared with the sham operation group, model group rats administered VEGF levels significantly increased, the difference was significant (P<0.01); Compared with model group, high dose group, middle dose group and Naoxintong group VEGF levelssignificantly increased, the difference was significant (P<0.01); rat VEGF levels increased in low-dose group, the difference was statistically significant (P<0.05).Compared with the low-dose group, high-dose group, middle-dose group and Naoxintong group rats, VEGF levels were increased, the difference was statistically significant (P <0.05).(3)Morphological changes of the brain tissue:Sham group:rats brain tissue with normal morphology, uniform staining of brain regions, a clear organizational structure and dense, granular nerve cells, cone-shaped and irregular in shape, cytoplasm pink, the nucleus blue dye, outline clear;Model: rat ischemic brain tissue structural disorder, cell dissolved, fuzzy nerve fibers loose, severe edema, visible phenomenon of perineural cells and foam cells, white matter structural damage.Low-dose group:ischemic brain tissue structural disorder, interstitial edema, pale staining, showing that the number of neurons decrease was degeneration, necrosis, glial cells relative increase in the phenomenon of visible perineural cells and foam cells, part of the blood vessels to dilateperivascular edema, visible inflammatory cell infiltration.Middle dose group and high dose group and Naoxintong group ischemic brain tissue districts can also see a change in the low-dose group, but the organizational structure disorder and interstitial edema, ischemic area decreased necrotic cells was significantly reduced, the degree of cell degeneration mitigate.(4)Each group of rats with cerebral ischemia microvessel density (CD31) results:Compared with the sham group, model group rat brain CD31expression was elevated, the difference was statistically significant (P<0.05) the groups administered rat brain tissue CD31expression was significantly higher, the difference was significant (P<0.01); compared with the model group, high dose group, middle dose group and Naoxintong group rat brain tissue CD31expression was elevated, and the difference was statistically significant (P<0.05), the low-dose group rat brain tissue CD31expression,increased, but not statistically significant (P>0.05); compared with the low-dose group, high dose group, middle dose group and Naoxintong group rat brain tissue increased expression of CD31, the difference was statistically significant (P<0.05); between the high-dose group, middle dose group and Naoxintong of group group pairwise comparison, rat brain tissue CD31expression was no significant difference(P>0.05).(5) Zhilonghuoxue tongyu capsule on cerebral ischemia TGF-beta1protein expression in rat brain tissue:Compared with the sham group, model group in rat brain tissue TGF-betal expression increased, the difference was statistically significant (P<0.05), administered rats brain tissue TGF-betal expression was significantly increased, the difference was significant(P<0.01); compared with model group, high dose group, middle dose group and the Naoxintong group rat brain tissue TGF-betal expression was increased, the difference was statistically significant (P<0.05), low-dose groupbrain tissue TGF-beta1expression was increased, but no significant difference (P>0.05); compared with the low-dose group, high dose group, middle dose group and Naoxintong group rat brain tissue expression of TGF-betal increased, the difference was statistically significant (P<0.05); pairwise comparison between the high-dose group, middle dose the group and Naoxintong group group, the brain tissue TGF-betal expression was no significant difference (P>0.05).(6)Zhilonghuoxuetongyu capsule on cerebral ischemia in rat brain tissue Ang-1, Tie-2protein expression:Compared with the sham group, model group Ang-1, Tie-2expression increased, the difference was statistically significant (P<0.05), the rats administered Ang-1, Tie-2expression was significantly increased,the difference was significant (P<0.01); compared with model group, low dose group rats Ang-1, Tie-2expression increased, the difference was statistically significant (P<0.05), the high-dose group, middle dose group andgroup of Naoxintong rat Ang-1, Tie-2expression was significantly increased, the difference was significant (P<0.01); compared with the low-dose group and high dose group, middle dose group and Naoxintong rats Ang-1, Tie-2expression increased, the difference was statistically significant (P<0.05); high dose group, middle dose group and Naoxintong rats Ang-1, Tie-2expression group pairwise comparison, there was nosignificant difference (P>0.05).Conclusion:(1)Zhilonghuoxuetongyu capsule can significantly improve symptoms of neurological deficit cerebral ischemia in rats.(2)Zhilonghuoxuetongyu capsule can increase cerebral infarction ischemic microvascular density, and promote recovery of neurological function, which may be to promote brain tissue TGF-β1, Ang-1, Tie-2protein expression by elevated serum VEGF levels,so as to promote cerebral infarction, ischemic area neovascularization, reduce neuronal apoptosis, reduce the leakage of the blood-brain barrier after cerebral ischemia, reduce cerebral edema(3).Zhilonghuoxuetongyu capsule in rats with focal cerebral microvascular protection with a certain dose-effect relationship.
