Objective:To establish the stable overexpression of Lumican in lungadenocarcinoma cell line A549and to investigate its effect to theproliferation of A549and its mechanism.Methods:The stable overexpression of Lumican in lung adenocarcinoma cellline A549was established. Furthermore, the cell growth curve,doublingtime,cell cycle and proliferation index was examined by MTT assay andflow cytometry. The status of the tumor in the experimental animals wasstudied. Finally,Western blotting was used to detect the expressions ofRhoC and p-Akt proteins in each group.Results:In Comparison with the other two groups, the growth of the cells inthe experimental group was faster (P <0.05), the doubling time wasshortened(P<0.05), the G1phase was shortened,while the S phase wasprolonged and the proliferation index went up(P<0.05), Subcutaneous tumors in volume and weigth increased and cells of the mitotic figuresbecame more (P<0.05), and the expressions of RhoC and p-Akt proteinswere increased (P<0.05), however,there was no significant differencebetween the control group and the empty vecor group (P>0.05).Conclusions:Lumican gene can promote the proliferation of lung adenocarcinomacell A549, and its mechanism may be related to increased RhoC and p-Aktproteins expression.
