Objective To assess the effect of KCNMA1transfected bonemarrow-mesenchymal stem cells (BMSCs) on the improvement of erectilefunction in diabetic rats.Methods The expression changes of KCNMA1were detected and potentialof multidirectional differentiation was measured before and after KCNMA1transfected into MSCs. Establishing the diabetes mellitus induced erectiledysfunction (DMED) rat model with streptozotocin (STZ) andapomorphine (APO). DMED rats were randomly divided into4groups: ratsunderwent intracavernous injection with PBS (DMED+PBS),non-transfected MSCs (DMED+MSCs), empty vector lentivirus transfectedMSCs (DMED+null-MSCs), and KCNMA1lentivirus transfected MSCs(DMED+KCNMA1-MSCs). Normal controls received intracavernousinjection of PBS (N+PBS). Erectile function was ascertained by measuringintracavernous pressure (ICP). The protein of KCNMA1was determinedwith immunohistochemistry in penile tissues.Results We transfected KCNMA1and blank vectors lentiviral successfullyinto the stem cells. The results of RT-PCR and Western blot showed thatKCNMA1over-expressed in the transfected KCNMA1cells comparedwith the blank vector cells and non-transfected cells. The lentivirustransfected MSCs had potential of multidirectional differentiation.3days after cells transplantation, green fluorescent cells were observed in variousareas of the penis;2weeks after cells transplantation, a part of the corpuscavernosum was observed to fluoresce green. The expression of KCNMA1in the corpus cavernosum was increased, and the DMED+KCNMA1-MSCsgroup displayed a significant improvement of erectile function.(P<0.05).Conclusion KCNMA1was able to enhance the positive effect of MSCs inthe treatment of diabetes associated erectile dysfunction.
