The formation of cervical cancer is long-term and multistep, and there are interaction elements in different stages of cervical lesions. As it is well-known, the persistent infection of human papillomavirus is the most important factor, that resulted in cervical cancer, among which HPV16is the most common one. HPV16gene combine with human gene, having influence on the cycle of host cells, and finally, it promotes fast grow of tumors. Hypoxia microenvironment and angiogenesis are necessary to the change of cervical cancer.We collected pathology department of115cases of blocks of cervical lesion tissue in the First Affiliated Hospital of Hebei Northern College, including45cases of SCC,30cases of CINⅡ,30cases of CINIII, and10cases of normal group. We detected the expression of HPV-16E6、HIF-1α and CD34by immunohistochemistry in tissue microarray to observe their relationship with cervical cancer progress, and detected C-MYC gene amplification by fluorescence in situ hybridization, make further discussion of the relationship between the early cervical neoplasia and molecular biology.Experimental results show, with the deterioration of the cervical lesions, positive rates of HPV-16E6and HIF-la were increasing, average values of MVD were ceaselessly aspiring. There is no significant difference in CIN groups, which of the expression of HPV-16E6(χ2=2.4434, P>0.05), others CIN group have significant difference(χ2=24.2458, P<0.01). There is no significant difference of the expression of HIF-la (χ2=0.6349, P>0.05) in normal group and CIN Ⅰ-Ⅱ group, others have significant difference(χ2=4.3981, P<0.05). The average values of MVD have significant difference in different CIN groups(P<0.05). The expression of HPV-16E6、HIF-la was correlate with the MVD in the cervical cancer tissue(r=0.55, P<0.05、r=0.31, P<0.05), also HPV-16E6was correlate with HIF-1α(r=0.42, P<0.05).The result of C-MYC gene amplification rate rises with the degree of cervical lesion(P<0.001), the positive rate was1.0%and36.6%,66.6%and77.7%in normal group and CIN I-II group, CINHIgroup and SCC group(r=0.624, P<0.001).In conclusion, HPV-16E6was correlate with HIF-la, maybe they have a synergistic effect in progress of cervical cancer. HPV-16E6can increase the expression and stability of HIF-la, lead new blood vessels to grow, and have important interaction in cervical cancer progress. C-MYC gene could function as an important biological indicator to forecast the risk level of cervical cancer, discriminating the cervical intraepithelial neoplasia with cervical cancer.
