Objectives: Survivin is a particular variety among the member of inhibitor ofapoptosis protein (IAP),which exists in nuclear and/or cytoplasm. The expression ofSurvivin protein is with tumour specificity, which only exhibits in the tumour andembryonic tissue and it is closely related to tumorigenesis, development, prognosis, cellproliferation, differentiation, invasion and metastasis. The research shows theexpression of Survivin gene is closely related to malignant grade of tumour and diseaseprognosis.Ovarian clear cell carcinoma is a kind of ovarian epithelial cancer whichoccupies5%-11%of ovarian cancer, most of patients are adult women, whose averageage is from48to58years old,10%of them are combined with hypercalcemia,25%-50%of them are combined with endometriosis. It is more unilateral and large. There isabundant vesicular cytoplasm containing abundant glycogen. There is deep-stainingnuclear with prominent atypia. It is easy to transfer to retroperitoneal lymph node andliver. It has not been seen the domestic research report about expression of Survivin inovarian clear cell carcinoma. Different expression of Survivin protein and gene betweenovarian clear cell carcinoma and normal ovarian tissue can be detected by this studywhich is to provide some research proof and treatment elements on molecule biologyfor diagnoses and treatment of ovarian clear cell carcinoma.Methods: This study was based on33patients who hospitalized and operated inDalian Obstetrics and Gynecology Hospital. The way of operation was staging ovariansurgery or cytoreductive surgery. Tumour tissue was fixed by formaldehyde andembeded by paraffin. The patients were approved with ovarian clear cell carcinoma bypathology result. Survivin protein expression was examined by immunohistochemistry(IHC) in ovarian clear cell carcinoma and normal ovarian tissue. The results wereanalyzed by Image-ProPlus6.0and SPSS18.0software. Total RNA was extracted fromformalin fixed paraffin embedded (FFPE) tissue. Survivin gene expression was examined by reverse transcription polymerase chain reaction(RT-PCR)in ovarian clearcell carcinoma(n=10) and normal ovarian tissue(n=6).Results: IHC experiment showed that expression of Survivin protein wassignificantly different in ovarian clear cell carcinoma and normal ovarian tissue. TotalRNA was successively extracted from FFPE tissue. RT-PCR showed geneticallyexpression of Survivin was significantly different in ovarian clear cell carcinoma andnormal ovarian tissue. Expression of Survivin protein and gene was related to stage ofcancer and size of focus. Had nothing to do with menopause or not and age of patient.Conclusions:The expression of Survivin protein and gene were significantlydifferent in ovarian clear cell carcinoma and normal ovarian tissue. It was possible thatSurvivin played a role in ovarian clear cell carcinoma development.
