Purpose: neuroblastoma cells cultured in vitro, the role of the PAP in neuroblastoma cells andby MTT assay, flow cytometry and RT-PCR analysis to analyze the PAP for neuroblastomacellproliferation and apoptosis, and in order to further explore the PAP to promote neuroblastoma cellproliferation and inhibition of apoptosis mechanisms laid the theoretical foundation for the nextexperiment.Method: In this study, neuroblastoma cells subcultured MTT assay, the role of the PAP inneuroblastoma cells, determination of its biological activity relationship analysis velvet antlerpolypeptides solution concentration and cell viability, which reveals itspromote neuroblastoma cellproliferation. By flow cytometry analysis of apoptosis of neuroblastoma cells and neuroblastomacells RNA extraction, PCR amplification, detection of Bcl-2, Bax expression by RT-PCR method,which reveals the PAP suppression neuroblastoma cell apoptosis mechanism.Results: The test proved by MTT cell activity and cell injury, polypeptides promoteneuroblastoma tumor cell proliferation effect, but also polypeptides having the biological activity,can be seen by the linear relationship of antler polypeptide solution concentration and cell viability,with antler polypeptide concentration of the solution increase, the cell viability was the growth ofthe state: when the antler polypeptides solution concentration in31.25μg/ml-125μg/ml betweencell survival index into a fast-rising stage. When the antler polypeptides solution doseconcentration125μg/ml about the cell viability index of relative peak values, when PAPconcentration between125μg/ml-250μg/ml, or when higher than250μg/ml, cell survival the ratefell into a plateau.PAP dose of800mg/ml apoptosis percentage of51.24%, can be observed400mg/ml and800mg/ml PAP PAP dose of400mg/ml, the apoptotic percentage is50.02%; neuroblastomatheapoptosis was significantly inhibited. Found by RT-PCR, the expression of Bax protein decreased,while the expression of Bcl-2expression continue to increase.Conclusion: PAP to promote neuroblastoma tumor cell proliferation effect is obvious, and thePAP has good biological activity.
