The Immunosuppression Of Hepatocellular Carcinoma Microenvironment Is Improved By Sini Decoction Through Interfering The Expression Of TLR4

Objective:This project is to study how Sini decoction improve the immunosuppression of Hepatocellular Carcinoma Microenvironment by interfering the expression of TLR4. Broadening the clinical idea for Fuzheng Treatment of Traditional Chinese Medicine on malignant tumor.Method:The inhibitions of Sini Decoction on proliferation of HCC cells were analyzed by MTT.HCC tumor models were established in mice by injecting of Hepal-6cells and the effect of Sini Decoction on tumor growths is also studied. CD4+CD25+Foxp3+regulatory T cells were detected by flow cytometry and the levels of IL-2、IL-6、IL-10、IFN-γ、TNF were measured by CBA.The expression of TLR4in liver cancer tissues was assessed by immunohistochemistry.Result:(1) Sini Decoction displayed a does-and time-dependent inhibition of the Hepal-6in vitro.(2) Sini Decoction could inhibit the growth of solid tumor in mice liver(P<0.05).(3)The weights and coefficients were increased after medium-dose、high-doses of Sini Decoction treatment.the weights and coefficients of spleens were increased after high-does of Sini Decoction treatment.(4) Sini Decoction decreased the proportion of CD4+CD25+Foxp3+regulatory T cells in serum、cancer tissues and para-carcinoma tissue.(5) Sini Decoction could increase the levels of IL-2、IFN-γ、TNF in serum、cancer tissues and para-carcinoma tissue of the HCC murine model.Could decrease the levels of IL-6、IL-10.(6) Sini Decoction could down-regulate the TLR4expression of cancer tissues.Conclusion:Sini Decoction could improve the the immunosuppression of Hepatocellular Carcinoma Microenvironment by increasing Thl cytokine、decreasing the Th2cytokine and decreasing the proportion of CD4+CD25+Foxp3+regulatory T cells.Down-regulating the TLR4expression maybe one of the mechanisms of anti-tumor effect.
