Objective:This study is to find out the endogenous mechanism of histoneacetylase p300with free radicals treated by moxibustion in myocardial cells after heavy exercise, and will be used to protect the heart during heavy sports training, enrich the theory of traditional Chinese medicine(TCM), also make TCM combine with sports medicine firmly.Methods:60adult ICR mice were randomly divided into blank control group (A), exercise control group (B), exercise and moxibustion therapy group (C)(n=20). Addition to the A, the other two groups swam without load every day for2weeks. In the first2days they swam30minutes, and gradually extended to1hour,l hour every time, once everyday, continued for two weeks.1hour after swimming training, the mice of exercise and moxibustion therapy group will be treated by moxibustion on ST36and RN4. Two weeks later, the activity of SOD and NOS, the content of MDA and NO in serum of the60mice were detected; the hearts of the mice were paraffin-embedded, sliced, and used to detect p300expression by the immunohisto-chemical methods.Results:(1) SOD activity:B Compared with A, the SOD activity was significantly decreased (P<0.01); C compared with B, the SOD activity was significantly higher (P<0.05).(2). MDA content:B Compared with the A, the MDA content was significantly higher (P<0.01); MDA decreased significantly when C compared with B (P<0.01).(3)NO content:B Compared with A, NO content was significantly higher (P<0.01); the content of NO of C was significantly higher than B (P<0.05).(4) NOS activity:B Compared with A, NOS activity was significantly increased (P<0.01); C was significantly higher than B (P<0.01).(5) p300expression:B Compared with A, there is no significant difference in the p300expression (P>0.05); there is a very highly significant difference between A and C, while B and C (P<0.01).Conclusion:Moxibustion can effectively remove free radicals in myocardial after heavy exercise by increasing the activity of SOD, reducing the content of MDA and control the concentration of NO. It may be related to HAT p300. HAT p300can activate the expression of the anti-apoptotic Bcl-2protein in myocardial cells.
