Objectives:To discuss the relationship between the telomere length in Beijing Han people’periphery blood leukocyte and human age; To explore the relationship of humanperipheral blood leukocyte telomere length and heart, kidney aging.Methods:Study object, whom from the target population participated in The People’Liberation Army General Hospital in2011, were divided into five groups according tothe age:35-44years,45-54years,55-64years,65-74years and>75years. Inaccordance with the strict inclusion and exclusion criteria, screened139cases ofhealthy people. The peripheral blood leukocyte telomere DNA restriction fragmentlength (TRF) was examined by Southern blotting with digoxigenin-labeledhybridization probe. The correlation between healthy human’s peripheral bloodleukocyte telomere length and the indexes which represent the heart, kidney functionwere also analyzed.Results:1.Correlation analysis were performed on the peripheral blood leukocyte telomerelength and Chronological age, the results showed the negative correlation, thecorrelation coefficient was-0.314(P<0.001) and the regression equation wasTFR=-0.033CA+8.269; The values of TRF from different age groups were analyzed byusing ANOVA, and the results showed telomere length in>75-year-old age group issignificantly shorter than the35-44,45-54and55-64age group(P <0.05). The valuesof TRF from different sexes were not different in different age groups (P>0.05).2.Correlation analysis and regression analysis were performed on the peripheral bloodleukocyte telomere length and renal function indexs, the results showed the negativeassociation between the telomere length and Cystatin C(r=-0.193, P <0.05), theregression equation was TFR=-2..020CYSC+7.827; The telomere length andglomerular filtration rate (GFR) did not show significant correlation(P>0.05).3. Correlation analysis and regression analysis were performed on the peripheral bloodleukocyte telomere length and cardiac function indexs, results showed that telomerelength and mitral E DT, the lower wall E and ascending aorta have correlation. thecorrelation coefficient were-0.216,0.225,-0.203respectively, and the regressionequation were TRF=-0.006(mitral E DT)+7.472, TRF=9.759(the lower wall E)+5.080,TRF=-0.082(ascending aorta)+8.783respectively.Conclusions:1. Negative correlation is shown between peripheral blood leukocyte telomerelength and Chronological age, telomere length shortened with the increasing age andtelomere length can be used as a biomarker of aging.2. The telomere length ofleukocyte is associated with CYSC, mean that telomere length has correlation withrenal function in elderly, may can as a predictor to reflect the change of kidney functionwith aging.3. The telomere length of leukocyte is associated with mitral E DT, thelower wall E and ascending aorta, may can as a predictor to reflect the change ofcardiac function with aging.
