OBJECTIVE:As oral glucose-lowering drugs which was found earliest and usedwidely, however, the question of whether sulfonylureas (SUs) increased thetumorigenesis risk in type2diabetes patients remained controversial. In order toevaluate the role of SUs on tumorigenesis in type2diabetes patients, a systematicreview of SUs therapy was performed.METHODS:Based on the principles and methods of Cochrane systematic reviews,electronic database searches was performed on the Cochrane library, PUBMED,EMBASE, Web of Science, CBM, CNKI, VIP, Wan fang, and the data from thebeginning of the database to January,2013were included. Language restrictions wereChinese and English. Randomized controlled trials and cohort study involved insulfonylureas therapy for type2diabetes mellitus were selected and assessed for themethodological quality, and the extracted data were performed meta-analysis bystatistical software Revman5.0RESULTS:7cohort studies and1clinical trial were eligible for meta-analysis. Aswas shown in meta-analysis,1) SUs therapy was significantly associated withincreasing risk of most, and especially prostate, pancreas, colorectal, lung, livercancer, but not in breast cancer, compared with metformin therapy.2) SUs therapywas significantly associated with increasing risk of most, and especially, colorectalcancer. Inversely, SUs therapy was associated with reducing risk in pancreas cancer,compared with insulin therapy.CONCLUSIONS: SUs therapy might increase the risk of most cancers. But reducingrisk in pancreas cancer, compared with insulin therapy. However, the validity of thisfinding requires supports of more large-size, prospective studies.
