Objective Investigating the feasibility of applying1H-NMR-based technique in the studies of plasma metabonomics of SD rats with diffuse axonal injury, and searching for the possible specific biomarkers to help early diagnosis of diffuse axonal injury.Methods The DAI animal model was established by using the modified Marmarou A method. All animals were randomly divided into5groups (control group (n=6), surgery one day control group (n=6), combat injury one day group (n=8), surgery three days control group (n=6), combat injury three days group (n=8)). After sacrificing all animals, collected the right ventricular blood and prepared the plasma for the1H-NMR detection, then use PCA, PLS-DA and OPLS-DA to process the data.Results The plasma1H-NMR of SD rat in DAI group was significant different from those in the control group.32kinds of molecule metabolites were significantly different between the plasma samples of each group. The Glu significantly increased in combat injury one day group, and the Lac significantly increased in combat injury three days group, while lipid composition (VLDL and LDL) significantly reduced in both combat injury one day and three days groups.Conclusions The1H-NMR is an effective technology to study the DAI metabolomics. Our analysis has identified32metabolites and their variations, which may provide reliable experimental basis for searching the specific biomarkers to help early diagnosis of DAI.
