Objective: To try to elucidate the chemosensitivity between two nasopharyngealcarcinoma cell lines and cisplatin.Method: The expression of RAD51in two nasopharyngeal carcinoma cell lines andnormal nasal epithelial cells was measured, and the expression of RAD51in CNE1and CNE2were analyzed by real time RT-PCR after treated with cisplatin; The cellsurvival rate of the CNE1and CNE2treated with cisplatin was also determined byMTT.Result: The mRNA levels of RAD51in CNE1cells and CNE2cells weresignificantly higher than those in normal nasal tissue cells (P <0.01); After treatedwith three doses of cisplatin(4μmol/L groupã€8μmol/L groupã€16μmol/Lgroup), the expression of RAD51in CNE2cells for4μmol/L group and8μmol/Lgroup were higher than CNE1cells (P <0.05); The cell survival rate in CNE1cells foreach dose group were lower than CNE2cells (P <0.01).Conclusion: Chemotherapy drugs to stimulate,low differentiated NPC cellsexpresses higher RAD51which decreases the chemotherapy sensitivity to cisplatin. |
