| Objective: To research the32kDa protein as a biomarker for schizophrene. Withprotein precipitation, protein electrophoresis, protein separation and mass spectrometry, toexplore the properties and the role of32kDa protein in clinical diagnosis.Methods: The samples of118serum (60with schizophrenia and58health people)were divided into two groups, PEG protein precipitation and trichloroacetic acid/acetoneprecipitation was used to obtain sediment samples, and selected a suitable method with bySDS-PAGE. The32kDa protein bands were cut and stored in-70℃. All the protein bandswere analyzed with LC-ESI-MSMS mass spectrometry.Results: The protein bands was not discovered by trichloroacetic acid/acetoneprecipitation in pre-tests, but were discovered by PEG protein precipitation which werestable and clearly. With7%PEG protein precipitation about a32kDa protein was foundedin schizophrenia patients (38/60), without in normal controls (0/58). The positive rate of32kDa protein band in schizophrenia is63.33%(38/60), and in normal controls was0%(0/58), with obvious statistical differences (P<0.01). With LC-ESI-MSMS massspectrometry,13proteins which the molecular weight is between30KD and35KD werefounded, among them6proteins had high expression. We also found3unknown proteins.Maybe these is the reason that abnormal protein band occurred.Conclusion: The schizophrenia patients have specific electrophoresis protein band.They are some unknown protein after mass spectrometry. |
