There is no satisfying treatment. The disease causes and pathogenesis is not clear. Sothe research on the pathogenesis of the disease, is the key to the research and development inthe treatment of UC specific drug. Chinese traditional medicine prevention and treatment ofUC, not only for UC pathogenesis, what is more important for its syndrome mechanism. Inrecent years, cytokines and other immunological mechanisms as well as a variety of signaltransduction pathways of research is a hotspot in the field of medical research. The subjectselected immunological compared classic cytokines IL-6and IL-10and the Mek in aRas/Raf/Mek/Erk signal pathway which has the less research involves in UC to study thehepatic stagnation and spleen deficiency type UC pathogenesis and intervention effect ofTongxieyaofang on the hepatic stagnation and spleen deficiency type UC.Objective: Study of Tongxieyaofang for repair of colonic mucosal injury induced byTNBS in experimental ulcerative colitis in rats and its mechanism.Methods: Prepare the UC models by means of the TNBS/ethanol enema and bindingmethod and intemperance of taking food, with high, medium and low three different doses ofTongxieyaofang intervention treatment of21days, and treated with SASP as positive controldrug. Therapeutic effect of Tongxieyaofang mainly through visual observation of colonicmorphology and microscope observation the pathological colon tissue of two score torepresent. IL-6, IL-10by enzyme linked immunosorbent assay (ELISA method) was used todetect the serum of experimental rats, at the same time using the method of RT-PCR detectionexperimental rats colon tissue Mek1and Mek2expression.Results: Compared with the blank control group, the content of IL-10in the model grouprat was decreased significantly(P<0.01), the content of IL-6in the model group rat wastransparently high(P<0.05).The content of IL-10in the each treatment groups wastransparently high and the content of IL-6was lower than the model group(P<0.05,P<0.01).Compared with the blank control group, the expression amount of Mek1/2mRNA of colonictissues of rats in the model group was decreased(P<0.01); after treatment, compared with themodel group, the expression amount of Mek1/2in the each treatment groups were increased, the discrepancy was statistically significant(P<0.01), especially the middle dose group andthe SASP group with the most obvious rising(P<0.01).Conclusion:Tongxieyaofang can be effectively treated by TNBS/ethanol enema andbinding method and intemperance of taking food experimental rat model of hepatic stagnationand spleen deficiency type ulcerative colitis was replicated symptoms.Tongxieyaofang can reduce the content of IL-6in serum of experimental rats, inhibit therelease of inflammatory factors, and increase the content of IL-10, promote theanti-inflammatory effect, the immune regulation of hepatic stagnation and spleen deficiencytype UC rats within the environment to achieve the effect of the treatment.The Tongxieyaofang can promote the relative expression amount of Mek1/2gene ofcolonic tissues of rats with UC. Effect of the Ras/Raf/Mek/Erk pathway in the regulationof hepatic stagnation and spleen deficiency type in UC, then realize the intervention effectof Shuganjianpi method. |
