Objective to research the effect of5FU plus the NSAIDs such as Celecoxibon the inhibition and invasion of Human colorectal carcinoma cells SW480.Methods the colorectal cancer SW480cell lines for the study, the experimentwere divided into four groups: the control (with DMSO alone), Celecoxib(50μmol/L),5Fu(40ã€10ã€1μmol/L), Celecoxib combine with5Fu(Celecoxib50μmol/L combinewith5Fu40ã€10ã€1μmol/L).The effect of5FU alone or in combination with celecoxibon Human colorectal carcinoma cells SW480were measured by MTT. The cellapoptosis rate was analyzed by flow cytometry. Transwell was used to measure theeffect of invasion of cell invasion between the monotherapy group and thecombination group. Western blot was used to detect the level of expression of Bcl-2ã€Bax in Human colorectal carcinoma cells SW480.Result1. It is found from MTT and flow cytometry: Celecoxib with5FU significantlyinhibited the growth of colorectal carcinoma cells and elevated the inhibition ratecompared with respective monotherapy group and the control group (P<0.05),andit is more obvious using low dose5FU(1μmol/L) in combination withcelecoxib(P<0.01).2. Transwell results show that the number of invasive cells in5FU(1μmol/L) andcelecoxib(50μmol/L)drug combination group was significantly higher than5FUmonotherapy group (P <0.05), the combination weakened colon cancer cellinvasion force. 3. Western blot analysis found that the combination group had more significantdecreased expression of Bcl-2and increased expression of Bax than monotheraphygroup.Conclution Celecoxib in combination with5FU can enhance5FU on the inhibitionof Human colorectal carcinoma cells SW480,and the promote apoptosis mechanismmay be related to poor expression of Bcl-2and high expression of Bax,resulting in thechange of intracellular Bcl-2and Bax. |
