ObjectiveAnalyse the single effect of environmental factors including personal living habits,family history, living history and so on between152couple of cases and controls, thepolymorphisms of gene IL6-572C/G, IL10-819A/G, IL10-592A/C, and the gene-gene,gene-environment interaction on the development of primary hepatic carcinoma (PHC), toprovide a reliable basis and preventive strategy for the follow-up studies.MethodsThis hospital based1:1matched case-control study was used for studying the riskfactors which associated with primary hepatic carcinoma.152pairs of cases and controlswere selected from the first people’s hospital of Shunde district. Using the molecularbiology techniques of TaqMan fluorescence quantitative PCR, analyse the polymorphismof IL6-572C/G, IL10-819A/G and IL10-592A/C, entered the data into EpiData3.0and usedSAS9.0software for univariate logistic analysis and multivariate analysis. Study theassociation between the polymorphism of IL6-572C/G, IL10-819A/G and IL10-592A/Cgene and the environmental factors to primary hepatic carcinoma; and study the interactionof gene-gene, gene-environment in the development of primary hepatic carcinoma. Results1Associated risk factors of primary hepatic carcinomaMultifactorial condition logistic final analysis results: drinking, eating raw fish, familyhistory of hepatitis B, family history of primary hepatic carcinoma and water pollutednearby home have statistical significance of association between them with primaryhepatic carcinoma, their respective OR is2.38(95%CI:1.13~5.00),2.43(95%CI:1.26~4.68),2.91(95%CI:1.40~6.03),3.75(95%CI:1.24~11.32),2.22(95%CI:1.17~4.20).2Interaction effects of IL6-572C/G, IL10-819A/G and IL10-592A/C polymorphismwith environmental factors to primary hepatic carcinomaThere is no interaction effect between IL6-572C/G polymorphism with environmentalfactors. In the interactive effect mode, smoking, drinking, eating raw fish, drinking ditchwater have a statistically significant associated with primary hepatic carcinoma.IL10-819A/G polymorphism have interaction effects with environmental factors, such assmoking and drinking ditch water. Their respective OR is14.13(95%CI:1.47~135.87),6.12(95%CI:1.01~36.89).In the interactive effect mode, drinking, eating raw fish, have astatistically significant associated with primary hepatic carcinoma.There is no interactioneffect between IL10-592A/C polymorphism with environmental factors. In the interactiveeffect mode, smoking, drinking, eating raw fish, have a statistically significant associatedwith primary hepatic carcinoma.Conclusions1Drinking, eating raw fish, family history of hepatitis B and PHC, water polluted nearbyhome have statistical significance of association with PHC, they may the main risk factorsof PHC in Shunde district.2In this study, there is no interaction effect between IL6-572C/G and IL10-592A/Cpolymorphism with environmental factors.IL10-819A/G polymorphism have interactioneffects with environmental factors, such as smoking and drinking ditch water.IL10-819A/Ggene polymorphism may be a molecular biological marker to the susceptibility of PHC.
