As a newly developed stationary phase material, penetrable silica demonstratedexcellent chromatographic properties. It possessed both mesopores and macropores atthe same time, which was in favor of large specific surface area, and ensured highcolumn efficiency, accelerated mass transfer and guaranteed low backpressure,respectively. In this thesis, penetrable silica was used as chromatographic matrix inbiopartitioning micellar chromatography (BMC), cell membrane chromatography(CMC) and cell membrane solid-phase extraction (CM-SPE) for profiling toxicity andtransdermal ability of benzophenone UV-filters and screening anti-tumor componentsin several Traditional Chinese Medicines.In the first chapter, BMC was used to establish QRARs models to profilebiotoxicity and transdermal ability of benzophenone UV-filters. In this chapter, theinfluence of concentration of brij35(0.01M,0.02M,0.03M) and pH of mobile phase(7.4and6.5) on the retention of benzophenone UV-filters on BMC were investigatedin detail. Multiple linear regression of QRARs was established between capacityfactor log K and log BCF and transdermal rate T under six chromatographicconditions. The R2of QRARs model between log K ang log BCF were0.9398~0.9753,while R2of QRARs model between log K and T were0.7569~0.8434. The resultsshowed that the penetrable C18based BMC could fast profile biotoxicity andtransdermal ability of benzophenone UV-filters.The second chapter also used BMC system to investigate the influence of brij35concentration (0.01,0.02,0.03,0.04M) and pH (6.5,7.4) of mobile phase on severalchemical drugs. Besides, their related parameters were used to establish QRARsmodels of oral pharmacokinetics parameters. The model was then used to predict oralphamacokinetics parameters of sildenafil and its similarities.In the third chapter, CMC and CM-SPE were used to screen underlyinganti-tumor components of several Traditional Chinese Medicine. Cell membrane ofHela cell was coated on the surface of penetrable silica for its large specific surface area. The new material was used for the packing of chromatographic column and SPEcolumn. Abundant epidermal growth factor on the surface of Hela cell was used toscreen anti-tumor components of Rhizoma curcumae, Angelica sinensis, hedyoticdiffusa, Rhizoma Coptidis, subprostrate sophora and herba patriniae.
