| Part One The function of TNF-a regulate the expression of connexin43in neonatal rat cardiomyocytesObjective:To discuss under the action of different concentration of TNF-α,the characteristics of cx43express in neonatal ratcardiomyocytes.Methods:Culture original generation of neonatal rat cardiomyocytes,then randomly divided the primary into four groups:TNF-α incubation groups, rhTNFR:Fc+TNF-a group,the normal group,. rhTNFR:Fc group.Culture primary neonatal rat cardiomyocytes48h,then the cardiomyocytes in the first group was cultivated with different concentration of TNF-α for24h,in the second group the cardiomyocytes was cultivated with rhTNFR:Fc and TNF-α for24h.The normal group without any treatment.The rhTNFR:Fc group was cultivated withrhTNFR:Fc. Western-blot and confocal were used to investigate the express changes of cx43effected by the TNF-α.Results:Compared with the normal group,the content of phosphorylation-cx43was decreased(p<0.05),and the content of the dephosphorylation-cx43was increased(p<0.05)in the TNF-α group.Compared with the TNF-αgroup,the degree of decreased expression of phosphorylation-cx43has been reduced.(p<0.05) And the increased degree of expression of the dephosphorylation-cx43has been reduced (p<0.05) in the rhTNFR:Fc+TNF-αgroup. In the rhTNFR:Fc group the expression of cx43close to normal group.Conclusions:Under the effects of TNF-α,the expression of phosphorylation-cx43has decreased,and the expression of dephosphorylation-cx43has increased in neonatal rat cardiomyocytes.And rhTNFR:Fc can reduce this role in a certain extent. Part two The mechanism of TNF-a effects the expression of cx43in neonatal rat cardiomyocytesObjective:TO discuss the mechanism of TNF-a effects the expression of cx43in neonatal rat cardiomyocytesMethods:Culture original generation of neonatal rat cardiomyocytes,then randomly divided the primary into four groups:normal group,TNF-α treatment group,TNF-α+SB203580treatment group,SB203580treatment group.Culture primary neonatal rat cardiomyocytes48h,then intervention the neonatal rat cardiomyocytes based on the above groups.Results:The decreased degree of express of phosphorylation-cx43has been reduced(p<0.05) and the increased degree of express of dephosphorylation-cx43has been reduced (p<0.05)in the SB203580+TNF-αgroup compared to TNF-αgroup.And there is no significant different ofcx43expression between SB203580group and normal group.Conclusions:It is possible that TNF-α can regulate the expression of cx43via p38mapk in neonatal rat cardiomyocytes.And p38mapk is an important Signaling pathway for TNF-α regulate the expression of cx43. |
