Naomaitong Formula consists of rhubarb, ginseng, kudzu root, Chuanxiong.Separately, the rhubarb plays the role of monarch of the prescription since it candetoxify toxin, improve blood circulation and remove blood stasis; and ginseng playsthe role of minister since it can benefit vital energy and strengthen the body resistance.While as a whole, the compatibility of all the drugs in it works in detoxifying toxin,descending turbidity, benefiting vital energy and strengthening the body resistance.The prescription treats effectively in cerebral infarction, and can significantly improvethe quality of life, together with the obvious protective effects on cerebral ischemiareperfusion injury.However, there is far less research on the material basis of the NaomaitongFormula, of which the effective substances are still not clear. Therefore, it is difficultto identify a quality control method which is in line with the traditional Chinesemedicine theory for Naomaitong Formula.Objective: By means of comparing the changes in chemical composition ofNaomaitong Formula in vivo and in vitro, investigate its transitional composition inblood, and explore the effective material basis of the Naomaitong Formula.Methods:1. Establish the HPLC fingerprint of Naomaitong Formula and conduct qualitycontrol of it, meanwhile, provide a suitable chromatographic condition toUPLC-TOF-MS.2. Use the established UPLC-TOF-MS chromatographic condition to analyze andidentify the chemical composition of Naomaitong Formula.3. Establish the UPLC-TOF-MS fingerprint with Naomaitong Formula in vivoserum to clarify the material basis of Naomaitong Formula via comparing the chemical composition changes both in vitro and in vivo.Results:1. Established the fingerprint of Naomaitong formula by HPLC. Twenty-onecommon peaks were identified, in which eight compounds were identified. At thesame time, the established fingerprint could effectively assess the quality ofNaomaitong formula, and also provided a reliable separation condition for theUPLC-TOF-MS.2. Established UPLC-TOF-MS Naomaitong formula total ion chromatogram,identified a total of thirty chemical constituents, and provided cross-references to theanalysis of transitional components in blood.3. Identified twenty transitional constitutions in blood through comparing thechemical composition changes both in vitro and in vivo, among which elevencompounds have already been identified. Those compounds have either prototypicalor metabolic components which are all likely to be the active substance basis ofNaomaitong formula.Conclusion:Comparing the chemical composition changes of Naomaitong formula both invitro and in vivo, to analyze and identify the transitional composition in blood, toclarify preliminarily the material basis of Naomaitong formula, and to provide areliable theoretical support for the optimization and quality control of Naomaitongformula’s prescription.
