Chapter1Reviewe of the genus of AconitumThis thesis is composed of three chapters: chapter1reviewed the distribution,chemical constituents and their biological activity of the genus of Aconitum. Chapter2focused on the purification and elucidation of chemical components from Aconitumbullatifolium.var.homotrichum W.T.Wang. Chapter3focused on the quality analysis of thecrude drug, preparation and setting up the initial standard for the drug.Chapter2Chemical composition of the Aconitum brachypodumDiterpenoid alkaloids is the dominating component of Aconitumbullatifolium.var.homotrichum W.T.Wang. Generally, its root was used as medicinal drugs. Thechemical constituent distinguishes depending on the place of origin.26compounds wereisolated from Aconitum bullatifolium.var.homotrichum W.T.Wang, which covered diterpene-alkaloids and other components. Anti-tumor activity and autonomic nerve activity wereevaluated for these compounds. As a result, songoramine were active in the vegetative nervebioassay.The root of Aconitum brachypodum is poisonous and used as an antipyretic orantipropagation in traditional medicine. Here we isolated one alkaloid, named songoramine,such structure is characterized by NMR assay, and very similar to phytohormone gibberellinacid, which can regulate growth and influence various developmental processes. Nitric oxideas the novel signal is also involved in multiple aspect of plant development. Here we found that songoramine at100nM can efficiently induce nitric oxide production, broke seedsdormancy and stimulate root growth in Arabidopsis thaliana, such effect could be reversed byNR or NOS-like inhibitor pretreatment to suppress NO release, or the Arabidopsis mutantdeficiency NO synthesis. Further results shower that Songoramine or NO treatment couldquickly accelerate GAI protein degradation, and enhanced PIN2protein accumulation, whileNO metabolism inhibitor treatment could delay GAI protein degradation and reduce PIN2protein accumulation. Basing on above results we propose that Songoramine as the novelalkaloid play the similar functional including breaking seeds dormancy and promoter rootelongation, likely phytohormone gibberelline, through NO siginal in plant.Chapter3Pharmaceutical analysis of Aconitum brachypodum PreparationThe quantities of Aconitum brachypodum were tested for drugs in order to explorethe possible way to strictly control the quality of drug of Aconitum brachypodum because ofthe complex species. Firstly, we have confirmed the high accuracy of the testing methodscreated by pharmacopoeia. And we have advanced some of the methods with the developmentof analytical chemistry. Secondly, we established fundamental understanding on the quality ofthe crude drug on the market. And analytical methods created for the quality test will bebeneficial to the quality regulation of the drugs. |
