| Objective:To study expression transformation of serum matrix metalloproteinase-9(MMP-9), organization MMP-9and E-cadherin detected in patients with breast cancerand evaluate its clinical significance in the breast cancer.Methods:The serum levels of MMP-9are measured by enzyme-linked immunosorbentassay(ELISA) based on serum extracted from20healthy women adults(Group a),20patients with cyclomastopathy (Group b),50patients with breast cancer (Group c)and50patients with breast cancer postoperatively (Group d). What’s more, we collect20normal breast tissue in patients with fiberoadenoma (Group A),20mammaryhyperplasia tissues (Group B) and50breast invasive ductal carcinoma tissues (GroupC). Immunohistochemistry is used to detect the expression transformation of MMP-9and E-cadherin, to analyze the relationship between MMP-9and E-cadherin in thedevelopment of breast cancer, to prove the correlation of MMP-9in serum and tissueof patients with breast cancer and additionally to analyze correlation between MMP-9and E-cadherin in breast cancer tissue.Results:1. MMP-9contents in serum of Group a, b, c and d are respectively95.26±2.65(ng/ml)ã€105.65±4.21(ng/ml)ã€212.53±3.83(ng/ml)ã€142.30±4.49(ng/ml).Compared to Group a, MMP-9in serum of Group c, d obviously addition, which hassignificant difference (P<0.05). Compared to Group a, MMP-9levels in Group b haveno significant difference (P>0.05). Compared to Group c, MMP-9content in Group dobviously decreases, which has significant difference (P<0.05).2. MMP-9in serum of Group c is independent of patient age and tumor size, whichhas no significant difference (P>0.05). However, it is related to histopathologygrading, tumor clinical staging and axillary lymph node metastasis, which has significance difference (P<0.05).3. The positive rates of MMP-9in Group A, B, C are respectively5.0%,30.0%,60.0%. Compared to Group A, the positive rate in Group B, C addition and hassignificant difference (P<0.05). Compared to Group B, the positive rate in Group Calso addition and has significant difference (P<0.05). MMP-9protein is related tobreast cancer histological grade, tumorous TNM staging and axillary lymph nodemetastasis, which has significant difference (P<0.05). However, MMP-9protein isindependent of patient age, tumor size, which has no significant difference (P>0.05).4. The positive rates of E-cadherin in Group A, B, C are respectively95.0%,85.0%and42.0%. Compared to Group A, the positive rate in Group B no significantdecreases(P>0.05). Compared to Group A, the positive rate in Group C obviouslydecreases, which has significant difference (P<0.05). Compared to Group B, thepositive rate in Group C obviously decreases, which has significant difference(P<0.05). E-cadherin is related to breast cancer histological grade, tumorous TNMstaging and axillary lymph node metastasis, which has significant difference (P<0.05).However, E-cadherin is independent to patient age and tumor size and has nosignificant difference (P>0.05).5. In33positive MMP-9samples of Group C, there exist only5positive E-cadherinsamples. However, we find13positive E-cadherin samples in the17negative MMP-9samples, which take a proportion of76.4%in positive expression. Based on SpearmanRank Correlation, we find that a significant negative correlation exists between theexpression of MMP-9and E-cadherin (r=0.605, P<0.05).6. There exists a positive correlation between MMP-9level in serum and MMP-9positive rate in cancer tissue of patient with breast cancer (r=0.886, P<0.05).Conclusion:1. MMP-9in serum, which might be closely related to carcinogenesis, progression,invasion and metastasis of breast cancer, has positive correlation to MMP-9positiverate in tissue. What’s more, MMP-9in serum has great help in distinguishing benign tumor and malignant tumor, evaluating curative effect of breast cancer operation andtrauma is less than the histology biopsy.2. MMP-9in breast cancer tissue and E-cadherin are both related to carcinogenesis,progression, invasion and metastasis of breast cancer and they both show significantnegative correlation in the expression of breast cancer. It is more valuable to detectthese two features together in the diagnosis of breast cancer. |
