| Objective: To investigate the effects of ω-3polyunsaturated fatty acids on kidney andinflammatory responses in burned rats.Methods: A total of72SD rats were randomly divided into3groups: the normalcontrol group (A group,n=8); the experimental group (B group,n=32); the treatmentgroup (C group,n=32).A group rats used warm water to simulate burn process, B groupand C group rats used30%total body surface area(TBSA) full thickness burns model.Rats in group A were injected NS (2mL/(kg·d)) by tail vein, Rats in group B wereinjected20%middle-long-chain fatty emulsion (2mL/(kg·d)) by tail vein, Rats ingroup C were injected ω-3fish oil (2mL/(kg·d)) by tail vein at the same time for eachday. Before injury group (Group A) and after injury groups (GroupB&C)12h,1d,4d,7d,10d were measured respectively the levels of serumPGE2ã€ET-1ã€IL-1ã€TNF-α; determined expression of kidney tissue TNF alpha mRNA by RT-PCRtechnology and observed morphological changes of kidney by normal light and TEM.Each subgroup had8rats.Results:1.Serum PGE2ã€ET-1ã€IL-1ã€TNF-αwere significantly higher than pre-injuryat each time point after injury in B&C,(P <0.01).. The levels in C group were lowerthan in B group, and were significantly different in4d,7d,10d after injury (P <0.05or P<0.01).2.Compared with Group A by TEM, Group B after injury nuclear membrane ofthe kidney tissue swelling,Mitochondria swelling and free bubble change,Crestbreakthrough bad;Group C after injury nuclear membrane of the kidney tissue swelling (compared with group B mitigation) or complete,Swelling of mitochondria (comparedwith group B mitigation),Crest breakthrough bad (compared with group B mitigation)or complete.3. Group B, group C at all time points after injury the expression of kidneytissue TNF-α mRNA was significantly higher than group A;Compared with group B,after injury,4d,7d, the expression of kidney tissue TNF-α mRNA significantly reducedin group C.Conclusions:1.ω-3PUFAs reduce the levels of serum PGE2ã€ET-1ã€IL-1ã€TNF-α.2. ω-3PUFAs reduce the expression of kidney tissue TNF-α mRNA.3. ω-3PUFAs reduce the damage of kidney after burn. |
