| Objective:To explore the effects of pressure therapy on cell cycle related gene P57kip2and CyclinE of hypertrophic scars on different stages. Along with observation of the proliferation and apoptosis of scar fibroblasts, we hope to provide a new theoretical basis for the pressure therapy of hypertrophic scars.Method:Scars harvest from surgeries included20treatment group cases and20control group cases. Also,5cases of normal skin were collected.The scar samples were divided into four groups according to the developing stages of hypertrophic scar: the3months,6months,12months and24months.The expression of P57kip2and CyclinE mRNA was detected by RT-PCR.At the same time detect PCNA (proliferating cell nuclear antigen) and apoptotic cells detected using TUNEL were counted, Changes in collagen of each groups using Masson law were observed statistical analysis was performed.Result:(1)mRNA expression of P57kip2:Following hypertrophic scar development,the expression mRNA of P57kip2in control group and treatment group from low level to high level.at3month,6month and12month,the mRNA expression of P57kip2of treatment group is higher than control group(P<0.05). While With no difference at24month between two groups(P>0.05).Inside each group,the expression intensity of mRNA P57kip2at3months was lower than that of6months(P<0.05).And that of3months and6months was lower respectively than that of12month,24month and normal group(P<0.05).In12month,24month and normal group were no difference (P>0.05).(2)CyclinE mRNA:mRNA expression of CyclinE in hypertrophic scar decreased with time in both treatment groups and control groups. At3month,6month and12month levels of mRNA expression of CyclinE was lower in treatment group than control group(P<0.05).With no difference at24month of two groups(P>0.05).inside each groups,the expression intensity at3months was higher than6months (P<0.05).And that at3months and6months expression was higher respectively than that of12month,24month and normal group(P<0.05). In12month,24month and normal group there were no difference(P>0.05).(3)PCNA expression:the expression of PCNA in hypertrophic scar all decreased with time in both control group and treatment group.Two groups inside,the expression intensity of PCNA were vary(P<0.05).At3month,6month and12month the expression of PCNA in two groups were difference(P<0.05).But with no difference at24month of two groups(P>0.05).(4)Apoptotic cell:the expression of apoptotic cells increased with time in both groups.inside each groups,the expression of apoptotic cell were vary(P<0.05).at6month,12month and24month the expression of apoptotic cell varied in two groups(P<0.05). With no difference at3month between two groups(P>0.05).(5)Collagen fiberrwith development of pressure therapy,the swirling collagen fiber bundle straightened,arranged loosely and orderly.Conclusion:(1)The abnormal expression of cell cycle regulatory gene P57kip2and CyclinE are closely related with the development of hypertrophic scar.(2)the expression of P57kip2and CyclinE shows that stress can accelerate the evolution progress of hypertrophic scar.(3)PCNA,apoptotic cell and collagen fiber detection from the treatment group shows that pressure can accelerate apoptosis and inhibition of cell proliferation,thereby inhibiting scar formation.(4)Pressure therapy can accelerate the evolution progress of the collagen fiber,making a swirl of collagen fiber bundle straightened,loosely and orderly arranged ones. |
