The Effect Of Chemotherapy Drug For Expression Of Livin And Caspase In The Glioma Cells

ObjectiveIn this vitro experiment, we detect the expression of livin and caspase-3-9、cell toxicity and the cell cycle in glioma U87MG cells after the intervention with five kinds of common tumor chemotherapy drugs.To study the diffterent roles of chemotherapy drugs and the apoptosis and anti-apoptosis pathway mechanism mediated livin and caspase-3-9,finding the ideas and methods to choose chemotherapeutics for clinical.MethodsWe use the logarithmic growth phase cells to expriment after using10kinds of different concentrations of cisplatin, Irinotecan,paclitaxel, carboplatin and5-fluorouracil. To detect the cell cycle by flow cytometry instrument drug intervention24h and test the cell proliferation with CTG kits,and then detect the expression of livin and caspase-3-9with RT-PCR after drug intervention72h.All datas was analysised by SPSS. Result.ResultsIn the glioma U87MG cells,the cells growth was inhibited stronger with increaseing of drug concentration of cisplatin, Irinotecan,paclitaxel, carboplatin and5-fluorouracil. The different inhibitory effect in the different drug in the same drug concentration and time.The cell cycle appeared significant change,increased in the G0/G1phase and reduced in S phase after drug intervention.The expression of livin more lower and the expression of caspase-3-9increaser than control group. The expression of livin and caspase-3-9were highly negative correlation. Conclusions1.The drug of cisplatin, Irinotecan,paclitaxel, carboplatin and5-fluorouracil have obvious inhibitory effect for U87MG cells.the inhibitory effect and drug concentration are positively correlation.2. The expression of livin was decreased and the expression of caspase-3-9was increased after drug intervention with five kinds of cisplatin, lrinotecan,paclitaxel, carboplatin and5-fluorouracil, The expression of livin and caspase-3-9were highly negative correlation.3. With different chemotherapy drug concentration increasing, The expression of livin was decreased and the expression of caspase-3-9was increased, The expression of livin and drug concentration was negative correlation,but the expression of caspase-3-9and drug concentration was positively related.4. The function of killing tumor cells was throuthed the expression of livin was inhibited lead to the expression of caspase-3-9increse after intervention with the chemotherapy drugs.5.1n the vitro study,the function of killing tumor cells in the five drugs,the effects of Irinotecan and paclitaxel are best, cisplatin and5-fluorouracil are better, carboplatin is weakest.
